摘要
试验旨在评估高脂肪日粮中添加不同水平的脂肪对小鼠脂肪代谢及氧化应激的改善作用。将初始体重为(22.79±1.75)g的100只小鼠随机分成5组,每组4个重复,每个重复5只小鼠。各组小鼠分别饲喂对照组日粮、高脂肪日粮、高脂肪日粮+20 mg/kg多糖、高脂肪日粮+40 mg/kg多糖和高脂肪日粮+80 mg/kg多糖,试验共开展30 d。结果:高脂肪日粮组小鼠试验结束后的体重较对照组显著提高了15.59%(P<0.05),日粮添加40和80 mg/kg多糖较高脂日粮组显著降低血清胆固醇、低密度脂蛋白、甘油三酯和血糖浓度(P<0.05)。此外,对照组、40和80 mg/kg多糖较高脂组和20 mg/kg多糖组显著提高了血清高密度脂蛋白浓度(P<0.05)。对照组、20、40和80 mg/kg多糖组较高脂日粮组显著提高了肝脏、肾脏和肺谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性(P<0.05),但丙二醛含量显著降低(P<0.05)。多糖处理组和对照组较高脂日粮组显著提高了肾脏和肺中维生素C和维生素E含量(P<0.05),但高脂日粮组肝脏维生素C和维生素E组含量显著高于其他组(P<0.05)。结论:高脂日粮中添加中药多糖在血脂代谢和血糖中起重要作用,同时可以防止小鼠机体自由基反应。
The aim of the trial was to assess the effect of adding different levels of fat to a high-fat diet on fat metabolism and oxidative stress in mice.The 100 mice with initial body weight of(22.79±1.75)g were randomly divided into 5 groups with 4 replicates in each group and 5 mice in each replicate.Mice in each group were fed a control diet,a high-fat diet,a highfat diet+20 mg/kg polysaccharide,a high-fat diet+40 mg/kg polysaccharide and a high-fat diet+80 mg/kg polysaccharide respectively for 30 days.Results:At the end of the experiment,the body weight of mice in the high-fat diet group was significantly increased by 15.59%compared with the control group(P<0.05),and the serum cholesterol,low-density lipoprotein,triglyceride and blood glucose concentration were significantly decreased in the high-fat diet group supplemented with 40 and 80 mg/kg polysaccharide(P<0.05).In addition,serum HDL concentrations were significantly increased in the control group,the 40 and 80 mg/kg polysaccharide high-fat group,and the 20 mg/kg polysaccharide group(P<0.05).The activities of glutathione peroxidase,superoxide dismutase and catalase in liver,kidney and lung were significantly increased in the control group and in the 20,40 and 80 mg/kg polysaccharide groups(P<0.05),but malondialdehyde content was significantly decreased(P<0.05).The contents of vitamin C and Vitamin E in the kidney and lung of the polysaccharide treatment group and the control group were significantly increased(P<0.05),but the contents of vitamin C and vitamin E in the liver of the high-fat diet group were significantly higher than those of the other groups(P<0.05).Conclusion:Polysaccharides added in high-fat diet play an important role in blood lipid metabolism and blood glucose,and can prevent free radical reaction in mice.
