摘要
本研究通过RNA-seq技术对3个长白山野杂猪与3个东北民猪背最长肌样本进行了mRNA测序和差异分析,从而为筛选长白山野杂猪肉质性状功能基因奠定基础。结果显示:在长白山野杂猪与东北民猪中均有100%以上的序列能完全比对到猪的基因组上,而所转录的基因占总基因的比例在3头长白山野杂猪中平均为59.67%,3头东北民猪为58.73%,而在不同个体间SNP位点数为14299~19671;在2个品种猪背最长肌中共表达基因有15842个,仅在长白山野杂猪中表达的基因有619个,仅在东北民猪中表达的基因有710个;在2个品种猪背最长肌中共获得显著差异表达的基因153个,其中在长白山野杂猪中上调表达的基因有55个,下调表达的基因有98个;GO分析显示,153个差异表达基因被注释到33条GO条目中,其中11条被注释到生物学过程中,11条被注释到细胞组分,11条被注释到分子功能;KEGG分析显示,这些差异表达基因被富集到130条通路中,其中主要与脂肪代谢相关的信号通路有关。结果表明,长白山野杂猪与东北民猪背最长肌中mRNA表达存在差异,这些差异表达基因将是将来研究2个品种猪间肉质差异的主要靶标之一。
mRNA sequencing and differential analysis of longissimus samples from 3 Changbai mountain semi-wild pigs and 3 Northeast min pigs were carried out by RNA-seq in order to lay the foundation for screening functional genes of meat quality traits in Changbaishan wild pig.in this study.Sequence alignment results showed that more than 100%of the sequences in the Changbai mountain semi-wild pigs and Northeast min pigs could be completely matched to the pig genome.And the proportion of the transcribed genes in the total gene was averagely 59.67%in the 3 Changbai mountain semi-wild pigs,and 58.73%in the 3 Northeast min pigs.The number of SNP loci among different individuals reached 14299~19671.There were total 17171 genes in longissimus dorsi of two kinds of pigs,including 619 genes expressed only in the Changbai mountain semi-wild pig,710 genes only in Northeast min pig;153 significantly differentially expressed genes were identified in the longissimus dorsi of two kinds of pigs,and 55 genes were up-regulated and 98 genes were down-regulated in Changbai mountain semi-wild pig;GO analysis showed that 153 differentially expressed genes were annotated to 33 GO entries,11 of which were annotated to the biological process,11 ones were annotated to the cellular components and 11 ones were annotated to the molecular function;KEGG analysis showed that these differentially expressed genes were enriched in 130 pathways,which were mainly related to the signaling pathway of fat metabolism.These differentially expressed genes will be one of the main targets for the future research on the meat quality differences among the two pig breeds.
