舒肝解郁胶囊对心肌梗死后抑郁大鼠行为学表现及心室电生理的影响

Influences of Shugan Jieyu Capsule on Behavior and Ventricular Electrophysiology in Depression Rats after Myocardial Infarction

目的研究舒肝解郁胶囊对心肌梗死后抑郁大鼠行为学及心室电生理的作用。方法通过结扎冠状动脉前降支制备急性心肌梗死模型以及慢性不可预见性温和应激建立抑郁模型,将65只SD大鼠随机分为5组:正常对照组(10只)、心梗组(15只)、抑郁组(10只)、心梗后抑郁组(15只)、舒肝解郁胶囊干预组(15只),应用糖水消耗试验及旷场试验评价行为学,酶联免疫吸附实验(ELASA)检测血浆去甲肾上腺素(NE)水平,心脏程序刺激测定梗死周边区及远离梗死区的有效不应期(VERP)及心室颤动阈值(VFT)。结果与正常对照组相比,心梗组、抑郁组及心梗后抑郁组糖水消耗百分比下降,运动量与直立次数减少,血浆NE水平、梗死周边区VERP增加,VFT下降,差异有统计学意义(P<0.05);舒肝解郁胶囊干预组上述行为学表现及血浆NE水平、VERP和VFT均较心梗后抑郁组改善(P<0.05)。结论舒肝解郁胶囊可通过调节心肌梗死后抑郁模型大鼠抑郁样行为及交感神经过度兴奋,改善心室电不稳定性。

Objective To observe the effect of Shugan Jieyu capsule on behavior and ventricular electrophysiology in depression rats after myocardial infarction.Methods Acute myocardial infarction model was prepared by ligating the anterior descending branch of coronary artery,and depression model was established by chronic unpredictability and mild stress.Sixty-five SD rats were randomly divided into 5 groups:normal control group(n=10),myocardial infarction group(n=15),depression group(n=10),post-myocardial infarction depression group(n=15),and Shugan Jieyu capsule group(n=15).The behavior was evaluated by sucrose water consumption test and open field test.Plasma norepinephrine(NE)level was dete cted by enzyme linked immune adsorption test(ELASA).The effective refractory period(VERP)in infarcted border zone and infarction remote zone,and ventricular fibrillation threshold(VFT)were detected by cardiac program stimulation.Results Compared with normal control group,the saccharine preference index,the amount of exercise,upright frequency,and VFT were decreased,the level of NE and VERP in infarcted border zone were increased in myocardial infarction group,depression group,and post-myocardial infarction depression group(all P<0.05).The above behavioral scores,plasma NE,VERP,and VFT were improved in Shugan Jieyu capsule group compared with that in post-myocardial infarction depression group,the differences were statistically significant(P<0.05).Conclusion Shugan Jieyu capsule may improve ventricular electrical instability by regulating depression-like behavior and sympathetic overexcitation in depression rats after myocardial infarction.
