成品与临用前配制氯化钾氯化钠注射液致痛作用比较及机制分析

摘要 目的比较成品与临用前配制氯化钾氯化钠注射液的致痛作用,并分析其可能的机制。方法将30只SPF级雄性昆明鼠随机均分为对照组、NK组及PK组各10只,分别给予腹腔注射生理盐水注射液、成品氯化钾氯化钠注射液(NK注射液)及临用前配制氯化钾氯化钠注射液(PK注射液)0.1 mL/10 g,观察15 min内出现扭体反应情况,ELISA法检测下丘脑组织中P物质(SP)、谷氨酸(Glu)含量;分组与用药同前,仅用药方法为足底皮下注射30μL/只,3 min内行疼痛行为学评分,免疫组化法检测脊髓L 4~L 6段组织中的SP、NK-1受体(NK1R)和Glu。结果PK组扭体反应发生率、疼痛评分,下丘脑组织中SP、Glu含量,以及脊髓组织中SP、NK1R、Glu表达量均高于对照组、NK组(P均<0.05);NK组与对照组各指标比较,差异均无统计学意义。结论NK注射液较PK注射液的致痛率和致痛程度降低,与其刺激所产生的中枢和外周疼痛神经递质减少有关。 Objective To compare the pain reaction on mice induced by two different potassium chloride and sodium chloride injection,and to analyze the probable mechanism.Methods Thirty male mice were randomLy divided into the normal saline injection(NS)control group,the finished 0.3%potassium chloride and sodium chloride injection(NK)group and the preparation of potassium chloride and sodium chloride injection 0.1 mL/10 g before use(PK)group,with 10 mice in each group.The occurrence rate of writhing reaction in 15 min was evaluated,and the content of substance P(SP)and glutamate(Glu)in the hypothalamus tissues was analyzed by ELISA.The grouping and medication were the same as before,we only changed the the medication method as the plantar subcutaneous injection(30μL/each).The algogenic effect in 3 min after subcutaneous injection was evaluated using pain behavioral scoring;then the content of SP,NK1R and Glu in the L4-L6 segments of spinal cord was analyzed by immunohistochemistry.Results The occurrence rate of writhing reaction,score of pain behavior,SP and Glu content in the hypothalamus tissues,and the expression of SP and Glu and NK1R in the spinal cord were significantly higher than those of the NK group(all P<0.05).There was no statistical significance in those indicators between NK group and normal control group(all P>0.05).Conclusion Compared with the PK injection,NK injection has lower algogenic effect by stimulating to produce less central and peripheral pain-related neurotransmitters.
