摘要
[目的]观察祛湿通络方含药血清对肺炎支原体(MP)感染的RAW264.7细胞肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)表达的影响。[方法]1)祛湿通络方含药血清的制备:SD大鼠随机分为空白对照组、中药低、中、高剂量组、阿奇霉素组,各组均连续给药7 d。2)MP感染的RAW264.7细胞炎症模型建立:模型组和各给药组分别加入MP菌液100μL,空白对照组加入100μL DMEM完全培养液,置于培养箱中培养2 h。3)酶联免疫吸附(ELISA)法检测祛湿通络方对MP感染的RAW264.7细胞TNF-α、IL-6表达的影响。[结果]与空白对照组比较,MP感染的RAW264.7细胞TNF-α、IL-6的分泌均显著上升(P<0.05);与模型组比较,各给药组对TNF-α、IL-6均有不同程度的下调作用(P<0.05),其中阿奇霉素组对TNF-α、IL-6的调节作用最优,中药低、中、高剂量组呈现剂量依赖性。[结论]祛湿通络方对MP感染的RAW264.7细胞TNF-α、IL-6的高表达具有下调作用,且呈剂量依赖性,这可能是祛湿通络方减轻炎症反应的机制之一。
[Objective]To observe the effect of Qushi Tongluo Formulation on TNF-αand IL-6 expression in RAW264.7 cells infected with mycoplasma pneumoniae(MP).[Methods]1)Preparation of drug serum containing Qushi Tongluo Formulation:SD rats were randomly divided into blank control group,low,medium and high dose group of traditional Chinese medicine,and azithromycin group.Each group received continuous administration for 7days.2)Establishment of MP-infected RAW264.7 cell inflammation model:model group and each treatment group were added with 100μL of MP bacterial fluid,and blank control group was added with 100μL of DMEM complete culture fluid,and cultured in incubator for 2 h.3)The effect of Qushi Tongluo Formulation on the expression of TNF-aand IL-6 in MP infected RAW264.7 cells was detected by ELISA.[Results]Compared with the blank control group,the secretion of TNF-αand IL-6 in MP-infected RAW264.7 cells increased significantly(P<0.05).Compared with the model group,TNF-αand IL-6 were down-regulated in all groups(P<0.05).[Conclusion]Qushi Tongluo Formulation can downregulate the high expression of TNF-αand IL-6 in MP infected RAW264.7 cells in a dose-dependent manner,which may be one of the mechanisms by which Qushi Tongluo Formulation can reduce inflammation.
