高效液相色谱法测定沾化冬枣中维生素C含量方法的优化 被引量:12

摘要 为建立一种测定沾化冬枣中维生素C(L-抗坏血酸)含量的高效液相色谱法,对提取方法及色谱条件等进行了改进优化。沾化冬枣中的维生素C采用1.0%的草酸溶液超声提取二次,以Kromasil C_(18)柱为分析柱,甲醇-0.1%草酸溶液(5:95,v/v)为流动相,在柱温为25℃,紫外检测器波长254 nm条件下检测。维生素C在20.0~100.0 mg/L范围内线性关系良好(R=0.999 9),相对标准偏差为0.30%(n=7),平均加标回收率为98.1%,检出限2.0×10^(-3) g/kg。结果证实本方法操作简单快捷,可以准确测定沾化冬枣中的维生素C。 In order to develop an optimal method for determination of vitamin C(L-Ascorbic acid)content in Zhanhua Winter-jujube by HPLC,extracting method and chromatographic conditions were improved.Vitamin C in the sample was extracted ultrasonically with 1.0%oxalic acid solution 2 times,Kromasil C 18 chromatographic column was used as stationary phase,the mobile phase was methanol-0.1%oxalic acid(5:95,V/V)with column temperature 25℃,and lastly detected at 254 nm through a UV detector.The standard working curve of vitamin C had a good liner relation in the concentration ranging from 20.0 to 100.0 mg/L(R=0.9999),RSD was 0.30%(n=7),the average recovery was 98.1%,and detection limit was 2.0×10-3 g/kg.The method was simple and fast,and could determine the content of vitamin C in Zhanhua Winter-jujube.
