PGAM5对食管癌细胞系增殖与转移能力的影响

摘要 目的体外实验探讨PGAM5基因对食管癌细胞系KYSE150增殖与转移能力的影响。方法通过Western Blotting法比较人正常食管上皮细胞系HEEC和人食管癌细胞系KYSE150中PGAM5蛋白表达水平;分别将对照siRNA(siCtrl)和PGAM5特异的siRNA(siPGAM5)导入KYSE150细胞系,Western Blotting法检测沉默PGAM5对抗凋亡基因Bcl-xL和迁移相关基因MMP2表达水平的影响;通过CCK8方法检测沉默PGAM5对细胞增殖的影响;流式细胞术检测对细胞凋亡的影响;划痕实验和Transwell实验检测对细胞迁移和侵袭能力的影响。结果 HEEC细胞系PGAM5蛋白表达水平显著低于KYSE150细胞系;siPGAM5转染KYSE150细胞系后Bcl-xL和MMP2蛋白表达水平显著降低;干扰PGAM5后KYSE150细胞的增殖能力显著降低,凋亡能力显著升高,同时细胞的迁移和侵袭能力显著降低。结论干扰PGAM5可以抑制人食管癌细胞系KYSE150的增殖和转移能力。 Objective To investigate the effect of PGAM5 on the proliferation and metastasis of esophageal tumor cell line KYSE150 in vitro. Methods Western Blotting was used to compare the PGAM5 protein expressions in both HEEC, a human esophageal epithelial cell line, and KYSE150, a human esophageal tumor cell line. siRNA(siCtrl)and siRNA(siPGAM5)were transferred into KYSE150 cell lines, respectively. Western Blotting was used to detected the expression level of Bcl-xL and MMP2;CCK8, flow cytometry assay, cell scratch and transwell assay were used to determine the cell proliferation, apoptosis, migration and invasion after the silencing PGAM5 of KYSE150 cell lines, respectively. Results The expression level of PGAM5 protein in HEEC cell line was significantly lower than that in KYSE150 cell line. The expression level of Bcl-xL and MMP2 proteins were statistically down-regulated by silencing PGAM5 in KYSE150 cell line. Transfected-siPGAM5 KYSE150 cells showed lower proliferation, migration, invasion and higher apoptosis. Conclusion The proliferation and metastasis of human esophageal tumor cell line KYSE150 were inhibited by silencing PGAM5 gene.
