摘要
目的:通过测定樟帮特色米泔水漂苍术的血清药物化学成分,发掘特色饮片,对比炮制前后差异,探讨其在大鼠体内的药效物质基础。方法:采用超高效液相-四极杆-飞行时间高分辨质谱法(UPLC-Q-TOF-MS/MS)技术,选择Waters Acquity UPLC?BEH C18(2.1mm×100mm,1.7μm)色谱柱,以乙腈-0.1%甲酸水溶液为流动相,在正离子模式下获得各成分裂解碎片信息,利用Peak View软件并结合文献对入血成分进行初步分析鉴定。结果:建立了米泔水漂苍术提取物血清药物成分的测定方法,共鉴定出7种成分,分别为酪氨酸、汉黄芩苷、苍术素、4-octylbenzoic acid、β-金合欢烯、β-桉叶醇和棕榈酸。结论:该方法准确、快速、有效、为明确樟帮特色米泔水漂苍术的体内药效物质基础提供了科学依据。
Objective:By measuring the chemical composition of the serum from rice-washed water bleaching Atractylodes Rhizoma,which has the zhangbang characteristics,to discover the special prepared drug in pieces and comparing before and after processing to explore the pharmacodynamic basis in rats in vivo.Methods:The chromatographic separation was performed on a Waters Acquity UPLC?BEH C18(2.1 mm×100 mm,1.7μm)with mobile phase composed of Acetonitrile and 0.1%formic acid solution and obtained fragmentation information of each component in a positivin vivoe ion mode,preliminary analysis and identification of blood absorption components by using Peak View software combined with literature.Results:A method for determining the serum drug composition of rice-washed water bleaching Atractylodes Rhizoma extract was established,and seven components were identified.These include Boc-D-Tyr-OH,β-D-Glucopyranosiduronic acid,Atractylodin,4-octylbenzoic acid,β-Farnesene,β-Eudesmol and Palmitic acid.Conclusion:The method is accurate,fast and effective and provides a scientific basis for clarifying the in vivo pharmacodynamics of rice-washed water bleaching Atractylodis Rhizoma which has the zhangbang characteristics.
