摘要
目的:观察青黛对脂多糖(LPS)诱导的人结肠上皮细胞(HCoEpiC)炎性细胞模型白细胞介素(IL)-6、IL-10的影响。方法:HCoEpiC根据给药血清的不同分为正常组,模型组,SASP组,青黛高、中、低剂量组,除正常组外均用LPS诱导造模,治疗24h后用ELISA、QPCR法测细胞中IL-6、IL-10蛋白及mRNA的表达。结果:青黛高剂量组IL-6蛋白表达较SASP组显著下降(P<0.01),与青黛中、低剂量比较,青黛高剂量组IL-6蛋白和mRNA表达显著降低(P<0.01)。青黛高、中剂量组IL-10蛋白表达较SASP组显著升高(P<0.01),且两者IL-10蛋白和mRNA表达较青黛低剂量组均显著升高(P<0.01,P<0.05)。结论:青黛能够抑制HCoEpiC细胞炎症模型促炎性因子IL-6的表达而增加抑炎性因子IL-10的表达,且青黛高剂量组的效果优于SASP组的作用,为该药的临床应用提供理论依据。
Objective:To observe the effects of Indigo Naturalis on the expression of inflammatory cytokines IL-6 and IL-10 in LPS-induced human colonic epithelial cells(HCoEpiC).Methods:HCoEpiC cells were divided into control group,model group,SASP group,Indigo Naturalishigh-dose group,Indigo Naturalis middle-dose group,and Indigo Naturalis low-dose group according to the different serum administration.Except the control group,the other groups were induced by LPS.And then the expressions of IL-6 and IL-10 protein and mRNA were measured by ELISA and QPCR after 24 hours of treatment.Results:Compared with SASP group,the level of IL-6 protein in Indigo Naturalis high-dose group decreased significantly(P<0.01).Compared with the Indigo Naturalis middle and low-dose groups,the expressions of IL-6 protein and mRNA of Indigo Naturalis high-dose group was significantly decreased(P<0.01).The expression of IL-10 protein increased significantly in Indigo Naturalis high and middl-dosee groups than that in SASP group(P<0.01),and both groups were better than the Indigo Naturalis low-dose group(P<0.01,P<0.05).Conclusion:Indigo Naturalis can significantly reduce the expression of pro-inflammatory factor IL-6 of LPS-induced HCoEpiC cells and increase the expression of anti-inflammatory factor IL-10.This experiment shows that the effect of Indigo Naturalis high-dose group is superior to that of SASP group,which provides a theoretical basis for the clinical application of the Indigo Naturalis.
