电针对睡眠剥夺大鼠学习记忆能力及海马区Rac1、Cdc42蛋白表达的影响 被引量:2

Effects of electro-acupuncture on learning and memory ability and expression of Rac1 and Cdc42 protein in hippocampus of sleep-deprived rats

摘要 目的:探讨电针对睡眠剥夺大鼠的空间记忆能力以及鼠脑海马区Rac1和Cdc42蛋白表达的影响。方法:将40只SPF级SD大鼠随机分为对照组、模型组、假电针组和电针组,每组10只;采用改良多平台水环境法制做大鼠睡眠剥夺模型。睡眠剥夺模型制作第1天起进行电针治疗,电针组电针"百会""大椎",断续波,频率2Hz,强度1mA;假电针组避开"百会""大椎"穴,在正常腧穴定位处向外旁开4mm,连接电针仪后不通电;电针和假电针治疗频次均为每日1次,每次15min,连续治疗5d。最后一次治疗结束后,进行Morris水迷宫测试。取大鼠海马,采用Western Blot检测Rac1、Cdc42蛋白的表达。结果:实验后电针组逃避潜伏期时间较模型组和假电针组下降,穿越平台次数较模型组和假电针组增加(P<0.05);与对照组比较,模型组Rac1、Cdc42表达均显著降低(P<0.05),电针组Rac1、Cdc42表达较模型组、假电针组显著增加(P<0.05)。结论:电针可调节突触可塑性相关蛋白表达,增强树棘突功能,改善睡眠剥夺大鼠的学习记忆能力。 Objective:To investigate the effects of electro-acupuncture on the spatial memory ability of sleep-deprived rats and the expression of Rac1 and Cdc42 proteins in the hippocampus of rats.Methods:40 SPF SD rats were randomly divided into control group,model group,sham electro-acupuncture group and electro-acupuncture group,with 10 rats in each group.Rat sleep deprivation model was established by modified multi-platform water environment method(MMPM).On the first day of making the sleep deprivation model,electro-acupuncture therapy was conducted.In the electro-acupuncture group,Baihui(DU20)and Dazhui(DU14)were given with intermittent wave,frequency of 2 Hz and intensity of 1 mA.In the sham electro-acupuncture group,avoid Baihui(DU20)and Dazhui(DU14)acupoints and open 4 mm outwards to the fixed points of normal acupoints.The treatment frequency of electro-acupuncture and sham electro-acupuncture was 1 time per day,each 15 min,continuous treatment for 5 d.After the final treatment,the Morris water maze test was performed.The expression of Rac1 and Cdc42 protein was detected by Western Blot.Results:Compared with model group and sham electro-acupuncture group,eluding latency time of electro-acupuncture group decreased and crossing platform times increased(P<0.05).Rac1 and Cdc42 were significantly lower in the model group than in the normal group(P<0.05),and significantly higher in the electro-acupuncture group than in the model group and the sham group(P<0.05).Conclusion:Electro-acupuncture can regulate the expression of synaptic plasticity related proteins,enhance the function of tree spines,and improve the learning and memory ability of sleep-deprived rats.
