基于PSD-95-NMDAR-nNOS通路研究针刺对单眼形觉剥夺大鼠视皮质神经细胞突触可塑性 被引量:4

Effects of acupuncture on synaptic plasticity of nerve cells in visual cortex of monocularly-deprived rats based on PSD-95-NMDAR-nNOS pathway

摘要 目的:研究针刺对单眼形觉剥夺大鼠视皮质神经细胞突触可塑性以及对NMDAR1表达调节的影响。方法:14日龄SD初生幼鼠30只,随机分为空白组、模型组、治疗组,各10只。除空白组外其余各组均右侧眼睑缝合制作形觉剥夺弱视模型。针刺治疗组大鼠右侧睛明、风池,大椎,隔日1次,每次10min,治疗30d。采用实时定量PCR技术,检测各组幼鼠视皮质NMDAR1、PSD-95、nNOS mRNA的相对表达量。结果:与空白组比较,模型组NMDAR1、PSD-95、nNOS mRNA表达显著降低(P<0.01),治疗组NMDR1 mRNA表达显著降低(P<0.05)。与模型组比较,治疗组NMDAR1、PSD-95、nNOS mRNA表达显著升高(P<0.01)。结论:针刺可以提高形觉剥夺性弱视大鼠视皮质NMDAR1、PSD-95、nNOS mRNA的表达,增加神经细胞突触后信号传导。 Objective:To study the effects of acupuncture on the synaptic plasticity and expression regulation of NMDAR1 in rats with monocularly-deprived rats.Methods:There were 30 newborn mice of SD age on the 14 th,randomly divided into control group,model group,and treatment group,each with 10.Except for the control group,the right eyelid suture was used to make amblyopia deprivation model.Treatment group rats right side bright,wind pool,large vertebrae,1 time every other day,10 minutes,30 days.Using real-time quantitative PCR(real-time PCR)technology,the relative expressions of NMDAR1,PSD-95,nNOS mRNA were measured in each group of young mice.Results:Compared with the control group,the expressions of NMDAR1,PSD-95,nNOS mRNA in the model group were significantly decreased(P<0.01),while NMDR1 mRNA in the treatment group were significantly decreased(P<0.05).Compared with the model group,the NMDAR1,PSD-95,nNOS mRNA expressions of the treatment group were significantly increased(P<0.01).Conclusion:Acupuncture can increase the expression of NMDAR1,PSD-95,nNOS mRNA in the visual cortex of rats with impaired vision,and increase the postsynaptic signal transmission of nerve cells.
