摘要
目的:研究化湿定痛汤对木瓜蛋白酶诱导的膝骨关节炎(KOA)的治疗作用,并探讨其作用机制。方法:40只SD大鼠随机分出16只空白组,另24只大鼠建立KOA大鼠模型,空白组和模型组各取8只比较,鉴定造模成功。将空白组和模型组大鼠各8只用0.9%氯化钠溶液灌胃、中药组大鼠8只用化湿定痛汤灌胃,给药4周取材。股骨髁、胫骨软骨进行HE染色,ELISA检测血清白细胞介素(IL)-1、IL-6、IL-10水平。结果:模型组Mankin’s评分显著高于空白组(P<0.05)。HE染色显示中药组软骨表层较模型组光滑,移行层较完整,细胞排列相对均匀。中药组IL-1、IL-6含量均显著低于模型组(P<0.05)、IL-10含量显著高于模型组(P<0.05)。结论:化湿定痛汤能在一定程度上延缓KOA大鼠软骨退化,这可能与其降低了KOA大鼠血清IL-1、IL-6水平及提升了血清IL-10水平有关。
Objective:To study the therapeutic effect of Huashi Dingtong Decoction on knee osteoarthritis induced by papain and explore its mechanism.Method:Forty SD rats were randomly divided into control group(n=16)and knee osteoarthritis model(n=24).Eight blank rats and eight model rats were compared to identify the success of the model.Eight rats in the blank group and the model group were given intragastric administration of normal saline and 8 rats in the traditional Chinese medicine group were given Huashi Dingtong Decoction for 4 weeks.HE staining was performed on femoral condyle and tibial cartilage,and serum IL-1,IL-6 and IL-10 levels were detected by ELISA.Results:The Mankin’s score of the model group was significantly higher than that of the blank group(P<0.05).HE staining showed that the surface articular cartilage in the blank group was smooth and smooth,the cells were arranged evenly and had no proliferation.In the model group,the tangential layer of cartilage was damaged,the fissures reached the bone radiation layer,and the cells gathered in clusters,which indicated that the model of knee osteoarthritis in rats was successful.The levels of IL-1 and IL-6 in the serum of Huashi Dingtong Decoction group were lower than those in the model group(P<0.05),and IL-10 in the serum of Huashi Dingtong Decoction group was higher than that in the model group(P<0.05).Conclusion:Huashi Dingtong Decoction can delay cartilage degradation in KOA rats,which is related to lowering serum IL-1 and IL-6 levels and increasing serum IL-10 levels in KOA rats.
