FERMT1在Stanford A型主动脉夹层患者中的表达及其临床意义 被引量:1

摘要 目的探讨主动脉夹层患者血管动脉壁中FERMT1的表达变化及其临床意义。方法13例急性Standford A型主动脉夹层患者主动脉血管标本作为夹层组,11例无主动脉夹层的心脏移植受体主动脉血管标本作为对照组。采用苏木精-伊红(HE)染色和弹性纤维(EVG)染色观察血管组织形态和弹性纤维的断裂情况;实时荧光定量PCR检测两组样本中FERMT1的mRNA表达;采用免疫组织化学染色和免疫印迹法检测FERMT1的蛋白表达水平;对主动脉直径与FERMT1 mRNA表达量行相关性分析。结果与对照组相比,夹层组动脉壁组织结构破坏,平滑肌细胞缺失,弹性纤维严重断裂;与对照组比较,夹层血管组织中FERMT1的mRNA含量显著降低(P<0.05),FERMT1的蛋白表达水平也显著降低(P<0.05)。此外,免疫组织化学染色结果显示FERMT1蛋白主要集中在血管中膜层细胞的胞质内。主动脉直径与FERMT1基因表达无显著相关性(P>0.05)。结论主动脉夹层患者动脉壁中FERMT1的mRNA和蛋白水平较对照组明显下调,该基因有可能作为未来预测和治疗主动脉夹层的新靶点。 Objective To investigate the clinical significance of FERMT1 expression in the vascular artery wall of patients with Stanford type A aortic dissection(TAAD).Methods Thirteen cases of TAAD patients were enrolled as the TAAD group and eleven cases of heart transplantation recipients results aortic dissection were enrolled as the control group.Hematoxylin-eosin(HE)staining and elastic Verhoeff's Van Gieson(EVG)staining were performed to evaluate morphological changes and rupture of elastic fibers of aorta.The mRNA expression levels of FERMT1 were verified by real-time PCR,and the protein levels of FERMT1 were analyzed by Western blot.Immunohistochemistry staining was conducted to observe the subcellular localization of FERMT1 in aorta of patients with aortic dissection.Finally,correlation analysis was performed to test relationship between inner aortic diameter and the expression of FERMT1 mRNA.Results Compared with the control group,HE staining and EVG staining showed aortic structure degeneration,loss of smooth muscle cells and rupture of elastic fiber.Compared with the control group,the mRNA levels of FERMT1 significantly decreased in TAAD group(P<0.05),and protein levels of FERMT1 also significantly reduced in TAAD patients(P<0.05).Immunohistochemistry staining showed that FERMT1 primarily located in cytoplasm.No significant correlation was found between aortic diameter and FERMT1 expression(P>0.05).Conclusions The mRNA and protein levels of FERMT1 in the arterial wall of patients with aortic dissection are significantly lower than that of the control group.FERMT1 may be a new potential target for the treatment and prognosis of aortic dissection in the future.
