基于网络药理学探讨威灵仙治疗痛风的作用机制

Mechanism of Clematidis Radix et Rhizoma in the Treatment of Gout Based on Network Pharmacology

目的:运用网络药理学方法探讨威灵仙治疗痛风的药理机制。方法:从Genecards、DisGeNET数据库中获取痛风的潜在靶点;从BATMAN-TCM数据库查找威灵仙活性成分及药物靶点;将药物靶点与疾病靶点进行映射,Cytoscape软件构建成分-靶点网络,分析网络拓扑参数(自由度和介数);将预测靶点导入David数据库,进行基因本体(GO)功能注释和京都基因与基因组百科全书(KEGG)通路富集分析,预测威灵仙治疗痛风信号通路。结果:预测得到威灵仙治疗痛风的24个有效成分和44个相关靶点,其中核心靶点是白细胞介素10(IL-10)、KCNQ1、维生素D受体(VDR)等,主要生物学过程包括一氧化氮生物合成正向调节、核因子κB(NF-κB)转录因子活性正向调节、炎症反应、免疫反应等,主要作用于细胞因子-细胞因子受体相互作用通路、肿瘤坏死因子(TNF)信号通路、过氧化物酶体增殖物激活受体(PPAR)信号通路等。结论:初步探讨了威灵仙治疗痛风的多成分、多靶点、多通路的作用特点,发现其可能通过影响免疫调节、炎症反应等发挥治疗作用。

Objective:To explore the pharmacological mechanism of Clematidis Radix et Rhizoma in the treatment of gout based on network pharmacology.Methods:The genes of gout were collected from Genecards and DisGeNET database,and the components of Clematis and their action targets were extracted from BATMAN-TCM database.The drug targets were mapped to disease targets,and then,the network of components and disease targets was established with Cytoscape software to analyze network topology parameters,such as degree and Betweenness centrality.Finally,the potential targets were imported into the David database to predict the main pathways.Results:Totally 24 active components of Clematidis Radix et Rhizoma and 44 drug targets were obtained.IL-10,KCNQ1 and VDR may be the core targets for the treatment of gout.The biological processes mainly included positive regulation of nitric oxide biosynthetic process,positive regulation of NF-κB transcription factor activity,inflammatory response and immune response,etc.Signaling pathways were related to Cytokine-cytokine receptor interaction,TNF signaling pathway,PPAR signaling pathway and so on.Conclusion:This study explored the multi-component,multi-target and multi-pathway of Clematidis Radix et Rhizoma in the treatment of gout,and found that it may play a therapeutic role by affecting inflammation and immune regulation.
