摘要
通过优化紫色红曲霉FDSJA-01的液态发酵生产工艺参数(搅拌器类型、溶氧、过程pH、补糖条件),以提高红曲色素的产量.优化后的5 L发酵罐液态深层发酵生产工艺条件为:采用六折叶DT602搅拌器,溶氧维持在(40±5)%,发酵pH值维持在(3.5±0.5),发酵96 h时开始以5 mL·h^-1的流速流加400 g·L^-1葡萄糖溶液,230 h时停止流加补糖.发酵264 h胞内色素含量(以每克湿菌体计算)达到最大值,红色素3454.9 U·g^-1,橙色素3746.7 U·g^-1,黄色素3295.8 U·g^-1,分别比优化前提高4.6、4.7、3.4倍,总色价提高4.2倍;发酵液桔霉素质量浓度为29.12μg·L^-1.
In this study,the production of monascus pigment was improved by optimizing the liquid fermentation process parameters(agitator type,dissolved oxygen,pH,and sugar-supplementing strategies)of Monascus purpus FDSJA-01.The results showed that the optimized conditions for the production of monascus pigment in 5 L fermentor were as follows:the fermentation agitator was the six-hinge stirrer DT602,the dissolved oxygen was maintained at(40±5)%,and the fermentation pH was maintained at(3.5±0.5).At the fermentation time of 96 h,400 g·L^-1 glucose was added at a rate of 5 mL·h^-1,and stopped at 230 h.At 264 h,the intracellular pigment content(calculated as per gram of wet cells)reached the maximum value.The content of red,orange and yellow pigment was 3454.9,3746.7 and 3295.8 U·g^-1 which was 4.6,4.7,3.4 times higher than before optimization,respectively.The total color value increased by 4.2 times and the the citrinin content was 29.12μg·L^-1.
