摘要
目的:探讨治疗功能性消化不良(FD)的作用机制。方法:将30只SD大鼠按随机数字表法分为空白对照组、模型组、电针组各10只。模型组及电针组采用多因素干预法造模20 d,空白对照组不给予任何处置。造模成功后电针组针刺足三里穴、太冲穴后接电针治疗14 d,余组不采用任何干预措施。采用ELISA(酶联免疫吸附剂测定)检测各组血清IL-6(白介素-6)、TNF-α(肿瘤坏死因子α)水平,WB(免疫印迹法)检测大鼠十二指肠TLR4(Toll样受体4)、NF-κB p65(核转录因子κB p65)表达。结果:与空白对照组比较,模型组大鼠十二指肠TLR4、NF-κB p65表达显著增高,血清IL-6、TNF-α水平显著升高;与模型组比较,电针组大鼠十二指肠TLR4、NF-κB p65及血清IL-6、TNF-α水平显著降低。结论:电针可能通过下调十二指肠TLR4、NF-κB p65表达,降低血清IL-6、TNF-α等炎性因子水平抑制炎症反应,发挥治疗FD作用。
Objective:To explore the mechanism of electro-acupuncture(EA)in the treatment of functional dyspepsia.Methods:A total of 30 rats were randomly divided into blank group,model group,and EA group with 10 rats in each group.Except for the blank group,multiple factor intervention methods were used to prepare the rat model with liver stagnation and spleen deficiency of functional dyspepsia.Then,EA therapy was been used for treating the molding groups for 20 days.Rats in blank group and model group were isolated in a silent environment during the treatment without any intervention.After treatment,Serum IL-6 and TNF-αlevels were detected by ELISA and the expressions of TLR4 and NF-B p65 were detected by Western blot.Results:the levels of TLR4,NF-κB p65 and serum IL-6 and TNF-αwere significantly higher in the model group than in the control group.Compared with the model group,the levels of TLR4,NF-κB p65 and serum IL-6 and TNF-αwere significantly decreased.Conclusion:EA may inhibit the inflammatory response by down-regulating the expression of TLR4 and NF-κB p65 in the duodenum and lowering the level of serum IL-6、TNF-αand other inflammatory factors,thus playing a role in the treatment of FD.
