基于网络药理学的防己茯苓汤治疗急性肾损伤机制研究 被引量:6

摘要 目的:利用网络药理学方法探讨防己茯苓汤治疗急性肾损伤的作用机制。方法:在网络药理学分析平台(TCMSP)检索防己茯苓汤中的化合物,经Swiss Target Prediction数据库获取其对应的靶点,运用Cytoscape3.7.1软件构建化合物-靶点网络并分析拓扑学,在CTD数据库筛选急性肾损伤的作用靶点,Venn分析化合物与疾病靶点,借助STRING数据库构建化合物与疾病靶点蛋白互作网络(PPI),最后在DAVID平台分析基因本体(GO)功能与KEGG通路富集。结果:共筛出防己茯苓汤127种化合物和273个靶点,急性肾损伤的靶点共496个,PPI共58个靶点,GO分析得到58个条目,KEGG分析得到62条信号通路。结论:本研究初步预测防己茯苓汤通过多靶点、多途径的药理作用机制治疗AKI,为深入研究其作用机制及临床运用提供新的思路。 Objective:Study on the Mechanism of Fangji Fuling decoction in the treatment of Acute Renal injury by Network Pharmacology.Methods:The compounds in Fangji Fuling decoction were searched on the network pharmacological analysis platform(TCMSP),and the corresponding targets were obtained by Swiss Target Prediction database.The compound-target network was constructed by Cytoscape3.7.1 software and the topology was analyzed.The target of acute renal injury was screened in CTD database,and the compounds and disease targets were analyzed by Venn.With the help of STRING database,the interaction network(PPI)between compounds and disease target proteins was constructed.Finally,the(GO)function of gene ontology and the enrichment of KEGG pathway were analyzed on the DAVID platform.Results:A total of 127 compounds and 273 targets of Fangji Fuling decoction were screened out,and 496 targets of acute renal injury were identified.There are 58 targets of PPI.58 items were obtained by GO analysis.62 signal pathways were obtained by KEGG analysis.Conclusion:This study preliminarily predicted that Fangji Fuling decoction through multi-target,multi-channel pharmacological action mechanism to play a role in the treatment of AKI,for in-depth study of its mechanism and clinical application to provide a new way of thinking.
