白术粗多糖的大孔树脂脱色工艺优化研究 被引量:11

摘要 采用大孔树脂对白术粗制多糖进行脱色,通过比较8种大孔树脂在脱色效果,筛选出HPD600树脂能较好地脱除多糖色素。进而以白术多糖的脱色率和多糖保留率为考察指标,通过单因素试验和正交试验对HPD600树脂脱色工艺进行优化。结果表明:在样品浓度25 mg/mL、样品pH6、脱色温度75 ℃、脱色时间6 h时,白术多糖脱色效果最好,脱色率和回收率可达79.75%和80.31%,表明大孔树脂法可有效地脱除白术多糖色素,为其进一步的分离纯化提供数据支撑。 Macroporous resin was used to decolorize the crude polysaccharide of Atractylodes macrocephala Koidz..By comparing the decolorization effect of 8 kinds of macroporous resins,HPD600 resin was selected to be able to decolorize polysaccharide pigment.Then,the decolorization process of HPD600 resin was optimized by single factor test and orthogonal test with decolorization rate and retention rate of Atractylodes macrocephala Koidz.polysaccharide as indexes.The results showed that the decolorization effect of Atractylodes macrocephala Koidz.polysaccharide was the best when the concentration of sample was 25 mg/mL,the pH was 6,the decolorization temperature was 75 ℃,and the decolorization time was 6 h.The decolorization rate and recovery rate were 79.75% and 80.31%.The results showed that macroporous resin method could effectively remove the pigment of Atractylodes macrocephala Koidz.polysaccharide,and provide data support for its further separation and purification.
