油茶皂苷提取物对HepG2细胞凋亡的影响 被引量:1

Effect of Saponin Extract from Camellia Oleifera on Apoptosis of Hep G2 Cells

摘要 探究油茶皂苷提取物对肝癌组织细胞(HepG2)的凋亡的影响,以进一步促进油茶皂苷资源的高值化利用。以油茶粕中油茶皂苷提取物为试材,以体外培养的HepG2细胞为对象。实验分为对照组、阳性对照组(30 μg/mL 5-氟尿嘧啶)、处理组(油茶皂苷5、30、50 μg/mL),以细胞存活率(凋亡率)和细胞内SOD、GSH-PX、MDA及其相关凋亡基因水平为综合评价指标,评价其抗癌效果的强弱。结果表明:当加入油茶皂苷对细胞处理12 h后,50 μg/mL浓度的油茶皂苷对细胞的存活率只有28.61%,较空白对照组降低了71.39%;此时细胞凋亡率为10.73,较模型组提高了157.93%。细胞内SOD水平(13.13 U/mg)、GSH-PX水平(44.03 U/mg)较对照组降低了64.40%、47.71%,且存在明显差异(P<0.05);而MDA含量显著升高,与对照组(1.28 nmol/mg)相比增加了201.20%。此时细胞Bcl-2、Cdkl、P13K、mTOR、VEGF基因的相对表达量较模型组降低了57%、65%、55%、37%、49%,细胞Bax、P53、caspase-3基因的相对表达量较模型组升高了51%、76%、74%。上述各指标均与阳性对照30 μg/mL 5-氟尿嘧啶处理组的效果基本一致。这说明油茶皂苷对HepG2具有较好的凋亡作用,且效果与常用抗癌药物5-氟尿嘧啶相近,可进一步将其开发成天然有效的抗癌产品并应用于市场中。 It studies the effect of camellia saponin extract on the apoptosis of hepatoma cells (HepG2) so as to further promote the high-value utilization of saponin resources of Camellia oleifera.In this paper,saponin extract of Camellia oleifera meal was used as a test material,and HepG2 cells cultured in vitro were used as experimental objects.The test was divided into a control group,a positive control group (30 μg/mL 5-fluorouracil),and a treatment group (camellia saponin 5,30,50 μg/mL).The cell survival rate (apoptosis rate),and intracellular SOD,GSH-PX,MDA and their related apoptotic genes were used as comprehensive evaluation index to evaluate their anti-cancer effect.The experimental results showed that when the camellia saponin was added to the cells for 12 hours,the cell survival rate of the camellia saponin at a concentration of 50 μg/mL was only 28.61%,which was 71.39% lower than that of the blank control group;at this time,the apoptosis rate was 10.73,which was an increase of 157.93% compared with the model group.The intracellular SOD level (13.13 U/mg) and GSH-PX level (44.0 U/mg) were reduced by 64.40% and 47.71% compared with the control group,and there were significant differences (P<0.05),and the content of MDA increased significantly compared with the control group (1.28 nmol/mg),which increased by 201.20%.At this time,the relative expression levels of Bcl-2,Cdkl,P13K,mTOR and VEGF genes in the cells were reduced by 57%,65%,55%,37% and 49% compared to the model group,and the relative expression of Bax,P53,and caspase-3 genes in the cells reduced by 51%,76% and 74% compared with the model group.The above indexes were basically consistent with the effect of the positive control 30 μg/mL 5-fluorouracil treatment group.This shows that camellia saponin has a better apoptotic effect on HepG2,and its effect is comparable to that of the commonly used anticancer drug 5-fluorouracil,which can be further developed into a corresponding natural and effective anticancer product and applied in the market.
