摘要
[目的]研究多发骨髓瘤(multiple myeloma,MM)患者骨髓液中微小RNA(miRNA,miR)-17-5p及磷酸酶和张力蛋白同源物(phosphatase and tensin homolog,PTEN)的表达及其临床意义。[方法]采用荧光实时定量PCR检测82例MM患者及40例健康人群骨髓组织中miR-17-5p及PTEN的表达,统计学分析两组间表达差异以及两者与临床病理特征之间关系。[结果]与对照组相比,MM组患者骨髓组织中miR-17-5p表达量明显较高,而PTEN表达量明显较低(t=15.365、11.647,P均<0.001)。MM组患者骨髓组织中miR-17-5p及PTEN表达与D-S分期、髓外浸润有关(t=9.713、3.147,8.901、2.259,P=0.000、0.002,0.000、0.027)。MM组患者骨髓组织中miR-17-5p与PTEN表达呈负相关(r=-0.611,P<0.001)。化疗前、后MM患者骨髓中miR-17-5p、PTEN的相对表达量无显著性差异(t=1.590、1.439,P=0.114、0.152)。化疗有效患者化疗后miR-17-5p表达较化疗前明显较低(t=2.149,P=0.034),而PTEN表达明显较高(t=2.252,P=0.026)。[结论] MM患者骨髓组织中miR-17-5p表达升高,而PTEN表达降低,两者与D-S分期、髓外浸润有关,有望成为MM诊断治疗的标志物。
[Objective] To study the expression of micro RNA-17-5 p(miR-17-5 p) and phosphatase and tensin homolog(PTEN)in bone marrow of patients with multiple myeloma(MM) and their clinical significance. [Methods] The expressions of miR-17-5 p and PTEN in bone marrow of 82 MM patients and 40 healthy subjects were detected by fluorescent real-time quantitative RT-PCR. The correlation of miR-17-5 p and PTEN expression with clinicopathological features of MM patients was analyzed. [Results] Compared with the control group,the expression of miR-17-5 p in bone marrow tissue of patients with MM was significantly higher,while the expression of PTEN was significantly lower(t=15.365 and 11.647,P<0.01). The expressions of miR-17-5 p and PTEN in bone marrow tissue of patients with MM were correlated with DS staging and extramedullary infiltration(t =9.713,3.147 and 8.901,2.259,P =0.000,0.002 and 0.000,0.027). There was a significant negative correlation between miR-17-5 p and PTEN expression in the bone marrow tissue of patients with MM(r=-0.611,P<0.001). There was no significant difference in the relative expression of miR-17-5 p and PTEN in the bone marrow of patients with MM before and after chemotherapy(t=1.590,1.439,P=0.114,0.152). After chemotherapy,miR-17-5 p expression was significantly lower(t =2.149,P =0.034),while PTEN expression was significantly higher(t =2.252,P =0.026) than those before chemotherapy in response patients. [Conclusion] The expression of miR-17-5 p in bone marrow tissue of MM patients is increased,while the expression of PTEN is decreased. Both of them are correlated with the D-S stage of MM and extramedullary infiltration,which indicates that miR-17-5 p and PTEN may be used as markers for diagnosis and treatment of multiple myeloma.
