不同类型猪肌肉组织中代谢及线粒体功能相关蛋白分析

Analysis of metabolism-and mitochondrial function-related proteins in muscle tissues in different types of pigs

摘要 为了研究影响家猪体型、肉质发育差异的因素,采用Western blot方法检测巴马猪、大白猪和姜曲海猪3种不同类型猪骨骼肌肌肉组织中线粒体功能相关蛋白细胞色素C氧化酶调节亚基4蛋白(COX4)、视神经萎缩症蛋白1(OPA1)、动力相关蛋白1(DRP1)、解偶联蛋白2(UCP2)以及与能量代谢相关蛋白B淋巴细胞瘤2蛋白(Bcl-2)、帕金森蛋白(Parkin)、Bcl-2相关X蛋白(BAX)的差异。结果显示:在线粒体相关蛋白比较检测中,4个蛋白均在大白猪中表达量最高,其次是巴马猪,在姜曲海猪中表达量最低,3组间差异均极显著(P<0.01);在能量代谢相关蛋白的比较检测中,Bcl-2/BAX蛋白表达量的比值在姜曲海猪(P<0.01)和大白猪(P<0.05)肌肉组织中极显著或显著高于巴马猪,Parkin蛋白在瘦肉型猪巴马猪和大白猪中均极显著高于脂肪型姜曲海猪(P<0.01)。综上,上述与促进或者稳定线粒体功能有关的蛋白均可能和促进猪肌肉生成,减少脂肪生成有关。 In order to study the factors influencing the differences in body size and meat quality of domestic pigs,Western blot was used to detect mitochondrial function-related proteins COX4,OPA1,DRP1 and UCP2,and energy metabolism-related proteins Bcl-2,Parkin,and BAX in the skeletal muscle tissues of three different types of pigs:Bama pigs,large white pigs,and Jiangquhai pigs.The results showed that,in the comparative detection,all the four mitochondrial-related proteins showed the highest expression in large white pigs,followed by the Bama pigs and the Jiangquhai pigs.The differences between the three groups were significant(P<0.01).In the comparative detection of energy metabolism-related proteins,the ratios of Bcl-2/BAX in the muscle tissues of the Jiangquhai pigs(P<0.01)and the large white pigs(P<0.05)were higher than that of the Bama pigs.Compared with that of the Jiangquhai pig,the Parkin protein was very significantly higher in the Bama pigs and the large white pigs(P<0.01).In summary,the above proteins might be associated with enhancing pig muscle production and reducing fat production.
