山羊痘病毒G9和A16基因的克隆及生物信息学分析 被引量:2

Cloning and bioinformatics analysis of the G9 and A16 genes of the goatpox virus

摘要 为研究山羊痘病毒(goatpox virus,GTPV)G9、A16蛋白的功能,用PCR方法从山羊痘病毒弱毒疫苗毒株GTPV AV41中扩增出编码G9、A16蛋白的基因进行测序,通过生物信息学方法对目的基因序列进行分析。结果显示:G9蛋白有34个磷酸化位点,17个抗原表位位点;二级结构中以α-螺旋和无规则卷曲为主,分别占36.61%和42.86%,有9个保守结构域,蛋白序列末尾有跨膜区域,包含15个半胱氨酸残基位点;三级结构中具有细胞凋亡和跨膜功能的结构。A16蛋白有36个磷酸化位点,19个抗原表位位点;二级结构中α-螺旋和无规则卷曲为主,分别占25.46%和48.28%,有10个保守结构域,蛋白序列末尾有跨膜区域,包含21个半胱氨酸残基位点;三级结构预测结果显示可以形成膜蛋白。上述研究结果为进一步探究GTPV的G9、A16蛋白功能提供了参考。 In order to determine the functions of the G9 and A16 proteins of the goatpox virus,PCR was used to amplify the genes encoding the two proteins from the GTPV AV41 strain of the attenuated goatpox virus vaccine for sequencing.Bioinformatics analysis was performed of the sequence of the target gene.The results showed that the GTPV AV41 G9 protein had 34 phosphorylation sites and 17 antigen epitope sites.Its secondary structure consisted mainly of an alpha helix and a random curl,accounting for 36.61%and 42.86%,respectively.It had 9 conservative structural domains,and at the end of sequence of the protein there was a transmembrane region containing 15 cysteine residue loci.The tertiary structure of the protein had apoptotic and transmembrane functions.The GTPV AV41 A16 protein had 36 phosphorylation sites and 19 epitope sites.In its secondary structure,alpha helix and irregular curl were dominant,accounting for 25.46%and 48.28%,respectively,and there were 10 conservative structural domains.At the end of the protein sequence,there were transmembrane regions containing 21 cysteine residue loci.The prediction results of the tertiary structure showed that membrane proteins might form.The above results laid a foundation for further exploring the functions of the GTPV AV41 G9 and A16 proteins.
