基于混池测序的犬恐惧行为相关变异位点的初步筛选

摘要 为了初步筛选与犬恐惧行为相关的变异位点,对6~8月龄的犬进行严格的恐惧行为测试,根据测试结果选取恐惧行为极端差异的史宾格犬20头,分为高恐惧行为组(FBH)和低恐惧行为组(FBL),每组各10头,采用混池测序的方法检测史宾格犬全基因组的单核苷酸多态性位点(SNP)和插入缺失位点(InDel),筛选与史宾格犬恐惧行为相关的变异位点。结果显示:通过特定条件的筛选,FBH组和FBL组存在差异的SNPs共14个,InDels共2个,这16个位点位于9个基因上。研究表明,通过混池测序的方法可以初步筛选出与犬恐惧行为相关的变异位点,进一步分析确定与恐惧行为遗传效应相关的变异位点,可以为犬遗传育种改良提供理论依据。 In order to preliminarily screen fear behavior-related mutation sites in dogs,20 Springer Spaniels aged 6 to 8 months with extreme differences in fear behavior were selected after a rigorous fear behavior test.These dogs were divided into the high fear behavior group(FBH)and the low fear behavior group(FBL),with 10 dogs in each group.Then,mixed pool sequencing was used to detect the sites of genome-wide SNPs and InDels in the dogs so as to screen the fear behavior-related mutation sites in the canines.The results showed that 14 SNPs and 2 InDels differing between the FBH and FBL groups were screened under specific screening conditions.These fear behavior-related mutation sites are located on nine genes.Analysis and identification of these sites for genetic effect might provide a theoretical basis for improvement in genetic breeding of canines.
