摘要
在单因素实验的基础上,以多糖提取率为响应值,以提取温度、提取时间、料液比为考察因素,采用响应面法优化甘草多糖提取工艺,并通过测定不同产地甘草多糖对DPPH自由基的清除率来评价其抗氧化活性。结果表明,甘草多糖的最佳提取工艺条件为:提取温度60.57℃、提取时间1.84 h、料液比1∶30.92(g∶mL),在此条件下,甘肃、内蒙古、新疆甘草多糖提取率分别为(19.89±0.08)%、(11.25±0.10)%、(9.60±0.13)%;当甘草多糖浓度为0.50 mg·mL^-1时,甘肃、内蒙古、新疆甘草多糖对DPPH自由基的清除率分别为27.17%、31.69%、58.68%。表明甘草多糖有一定的抗氧化活性。
Using the extraction rate of Glycyrrhiza polysaccharides as an evaluation index,we investigated the effects of extraction temperature,extraction time,and solid-liquid ratio on the extraction rate of Glycyrrhiza polysaccharides by response surface methodologies on the basis of single-factor experiments.Moreover,we evaluated the antioxidant properties of Glycyrrhiza polysaccharides from different producing areas by the determination of scavenging rate of DPPH free radicals.The results show that the optimum extraction conditions can be determined as follows:the extraction temperature of 60.57℃,the extraction time of 1.84 h,and the solid-liquid ratio of 1∶30.92(g∶mL).Under above conditions,the extraction rates of Glycyrrhiza polysaccharides from Gansu,Inner Mongolia,and Xinjiang reach(19.89±0.08)%,(11.25±0.10)%,and(9.60±0.13)%,respectively.When the concentration of Glycyrrhiza polysaccharides is 0.50 mg·mL^-1,the scavenging rates of DPPH free radicals by Glycyrrhiza polysaccharides from Gansu,Inner Mongolia,and Xinjiang reach 27.17%,31.69%,and 58.68%,respectively,revealing that the Glycyrrhiza polysaccharides have certain antioxidant activity.
