龙胆多糖的果胶酶法提取工艺及抗氧化活性研究 被引量:5

Optimization in Pectinase-Assisted Extraction Process of Polysaccharides from Gentiana scabra and Their Antioxidant Activity

摘要 以龙胆多糖提取率为评价指标,采用单因素实验和响应面实验优化龙胆多糖的果胶酶法提取工艺,并通过测定龙胆多糖对DPPH自由基和羟基自由基的清除率来评价其抗氧化活性。结果表明,龙胆多糖的最佳提取工艺为:果胶酶用量4%(以龙胆草质量计)、料液比1∶40(g∶mL)、提取温度62.98℃、提取时间5.25 h、pH值1,在此条件下,龙胆多糖提取率达到17.69%;龙胆多糖具有较好的抗氧化活性,且其抗氧化活性与浓度存在一定的量效关系。 Using the extraction rate of polysaccharides from Gentiana scabra as an evaluation index,we optimized the pectinase-assisted extraction process of polysaccharides from Gentiana scabra by single-factor experiments and response surface methodologies,and evaluated the antioxidant activity of polysaccharides from Gentiana scabra by the determination of the scavenging rates of DPPH free radicals and hydroxyl free radicals.The results show that the optimal extraction process is determined as follows:the pectinase dosage of 4%of Gentiana scabra,the solid-liquid ratio of 1∶40(g∶mL),the extraction temperature of 62.98℃,the extraction time of 5.25 h,and the pH value of 1.Under above conditions,the extraction rate of polysaccharides from Gentiana scabra reaches 17.69%.The polysaccharides from Gentiana scabra have strong antioxidant activity,and there is a dose-effect relationship between antioxidant activity and concentration.
