摘要
甲基转移酶3(methyltransferase-like 3,METTL3)是m^6A甲基转移酶复合体中的关键蛋白,能够催化RNA上的腺嘌呤(A)第6位氮原子发生甲基化。METTL3介导的m^6A修饰与RNA的剪接、出核、降解和翻译都有着密切联系,并且在肿瘤的发生发展中起着重要作用。该文主要介绍了METTL3的结构,以及其对mRNA翻译的调控,与非编码RNA的相互作用等多种生物学功能在肿瘤发生发展中作用,旨在为基础及临床医学中对m^6A修饰的研究提供新的研究思路和治疗靶点。
Methyltransferase-like 3(METTL3)contributes directly to the chemical modification of mRNA in eukaryotes by catalyzing methylation of adenine’s sixth nitrogen atom on RNA.METTL3-mediated m^6A modification exerts critical influences on RNA splicing,nuclear export,degradation and translation.Moreover,it also plays an important role in tumorigenesis.In this review,we discuss the structure of METTL3,and how its biological functions such as regulating mRNA translation and its interaction with non-coding RNA contribute to tumorigenesis and tumor progression,attempting to provide new ideas and therapeutic targets for further in-depth research of m^6A in basic and clinical medicine.
