梨黑斑链格孢菌次生代谢产物的分离鉴定及毒素含量分析 被引量:3

Isolation and Identification of Secondary Metabolites from Alternaria Gaisen as Well as Biotoxins Content Analysis

摘要 通过分离鉴定梨黑斑链格孢菌次生代谢产物,比较分析梨链格孢菌利用库尔勒香梨、苹果、西红柿、马铃薯、大米、燕麦等不同营养基质后的毒素含量。采用硅胶柱色谱、制备高效液相色谱、1D&2D NMR技术等方法,分离鉴定梨黑斑链格孢菌次生代谢产物,高效液相色谱法分析毒素含量。从梨链格孢菌中分离鉴定了6个次生代谢产物,分别为交链孢烯(1)、链格苝醇(2)、交链格孢酚(3)、交链格孢酚单甲醚(4)、土曲霉酮(5)和7-氨基-1,3-二羟基-苯丙酮(6);发现梨链格孢菌利用不同营养基质所产生物毒素(化合物1-4)结构种类相同,但含量存在显著差异。对梨链格孢菌生物毒素成分进行了定性定量分析,明确了毒素成分的主要结构,揭示了该菌利用不同营养基质所产生物毒素含量差异的特点。 Through the separation and identification of the secondary metabolites of Alternaria gaisen,compare and analyze the differ⁃ence of toxin producing of this fungus using Korla pear,apple,tomato,potato,rice and oat as nutritional substrates.Silica gel column chromatography,prepared high-performance liquid chromatography,and 1D&2D NMR technologies were used to determine the struc⁃ture of secondary metabolites produced by A.gaisen,and HPLC method utilized to analyze the content of biotoxins.Six compounds were isolated and identified as alternuene(1),altenuisol(2),alternariol(3),alternariol methyl ether(4),terrein(5)and 7-amino-1,3-dihy⁃droxy-propiophen(6)from A.gaisen,respectively.The structrue types of biotoxins produced by A.gaisen utilizing different nutritional substrates weresamewhile obvious differences in the content of biotoxins were observed.Based on the qualitatively and quantitatively analyses of biotoxins from A.gaisen,the structure of main biotoxins was clarified and the content difference of biotoxins from A.gaisen with different nutrition illustrated.
