基于RNA-seq的水稻根尖和侧根区愈伤组织诱导中生长素相关基因差异表达分析 被引量:3

RNA-seq-based differential expression analysis of auxin-related genes during callus induction derived from root tip and lateral root region in rice(Oryza sativa)

摘要 本研究以愈伤诱导培养基(CIM)诱导24 h的水稻(Oryza sativa)主根为材料,利用高通量转录组测序(RNAseq)技术进行主根根尖和侧根区的基因差异表达分析。通过对生长素信号相关基因家族进行聚类和差异表达分析后发现:生长素受体基因OsTIR/AFBs及其同源基因在根尖和侧根区组织的愈伤诱导过程中均无明显的差异表达;系统进化关系接近的OsIAA20、OsIAA9等10个OsIAA基因在根尖和侧根区均增强表达;而另一组进化关系相邻的Os IAA11和Os IAA13基因在根尖和侧根区表现出相反的差异表达模式。相比而言,Os ARFs家族基因在根尖和侧根区中对生长素诱导的表达变化具有明显的差异性。本研究分析了在根尖和侧根区组织生长素信号相关家族基因差异表达的异同,拟为根愈伤组织诱导中生长素响应的组织特异性研究提供新参考。 In this study,we used high-throughput transcriptome sequencing(RNA-seq)to analyze differentially expressed genes in root tip(RT)and lateral root region(LRR)when rice(Oryza sativa)primary roots were induced on callus induction medium(CIM)for 24 hours.Through phylogenetic relationship and differential expression analyses of gene families involved in auxin signaling pathway,we found that the expressions of auxin receptor OsTIR1/AFBs family genes did not apparently change during callus induction in RT and LRR.The expressions of 10 OsIAA genes with close phylogenetic clades,including OsIAA20 and OsIAA9,were enhanced dramatically in RT and LRR.However,a clade of OsIAA11 and OsIAA13 with adjacent evolutionary relationship showed an opposite differential expression pattern between RT and LRR.In contrast,the differential expressions of OsARF family genes were apparently different between RT and LRR.This study analyzed similarities and differences of the differentially regulated expression of auxin-related genes between RT and LRR,providing valuable data for further investigating the tissue specificity of auxin response in the root-derived callus induction.
