摘要
目的:探讨二甲醚吸入染毒对雌性大鼠卵巢和生殖激素的影响。方法:将18只雌性SD大鼠随机分为对照组、低剂量组和高剂量组,每组6只。对照组大鼠吸入空气,染毒组大鼠动式吸入含二甲醚的空气,二甲醚浓度分别为0.5%和1.0%(体积分数),2 h/d,5 d/周,连续染毒4周。染毒结束后测定大鼠血清中生殖激素的水平,计算卵巢脏器系数,进行卵巢和子宫组织病理学观察。结果:与对照组比较,两组大鼠的体重、卵巢重量和卵巢脏器系数均未发生明显变化(P>0.05),卵巢和子宫未见明显病理损伤;高剂量组雌性大鼠孕酮明显升高,差异有统计学意义(P<0.05);黄体生成素和抑制素B水平明显降低,差异均有统计学意义(P<0.05);低剂量组未见明显变化。结论:亚急性吸入二甲醚会引起雌性大鼠生殖激素水平紊乱。
Objective:To explore the effects of dimethyl ether(DME)inhalation exposure on ovarium and reproductive hormones of female rats.Methods:Eighteen adult female Sprague Dawley rats were randomized into 3 groups(n=6 for each).Two groups were exposed to DME in air in different dosage(0.5%and 1.0%v/v,respectively),2 h/d,5 d/week for 4 weeks in a dynamic inhalation chamber system.Another group served as control was treated with normal air.After exposure,serum,ovarium and uterus were harvested for determination of serum reproductive hormone levels,organ coefficients of ovarium and histological changes of ovarium and uterus.Results:Compared with the control group,there were no significant difference in the body weight,ovarium weight and organ coefficients of ovarium in the treated groups than those in the control group(P>0.05).No histological abnormalities were observed in ovarium and uterus.Rats treated with higher DME showed significant increase in the levels of serum progesterone(PROG)(P<0.05),significant decrease in the level of serum luteinizing hormone(LH)and inhibin B(INH-B)(P<0.05).No significantly difference in the level of serum reproductive hormone were observed in low-dose group in comparison with control(P>0.05).Conclusion:Sub-acute inhalation of DME may induce an imbalance of sex hormones in female SD rats.
