摘要
目的探讨酪蛋白磷酸肽-纳米无定形磷酸钙(CPP-ACP)在三聚磷酸钠(TPP)辅助下对脱矿牙本质片再矿化的影响。方法制备脱矿牙本质片实验模型并进行再矿化处理21 d,根据处理方式不同分为CPP-ACP再矿化组(A组)、CPP-ACP+TPP联合再矿化组(B组)、单独模拟唾液再矿化组(C组)、阴性对照组(D组)以及阳性对照组(E组),通过衰减全反射-傅里叶变换红外光谱(ATR-FTIR)、扫描电镜(SEM)、透射电镜(TEM)以及X射线能谱分析(EDS)等方法评价各组的再矿化效果。结果ATR-FTIR检测结果显示,CPP-ACP在TPP辅助再矿化21 d的脱矿牙本质样片与天然牙本质样片的红外图谱的峰位置及峰强度大致拟合;B组可以在脱矿的牙本质表面均匀沉积再矿化物,完成牙本质小管的封闭,且牙本质胶原纤维内可见周期样“横纹”结构;B组再矿化后牙本质样片表面钙/磷元素摩尔比显著高于A、C、D组,差异有显著性(F=19.27,P<0.05);B组与E组比较差异无显著性(P>0.05)。结论CPP-ACP在TPP辅助下对脱矿牙本质片可以产生较理想的仿生再矿化效果。
Objective To explore the effect of casein phosphopeptide-amorphous calcium phosphate(CPP-ACP)on the remineralization of demineralized dentin slices with the assistance of sodium tripolyphosphate(TPP).Methods An experimental model of demineralized dentin slices was prepared,and remineralization was carried out for 21 d.According to the method of remineralization,they were divided into CPP-ACP remineralization group(group A),CPP-ACP+TPP combined remineralization group(group B),simulated saliva remineralization group(group C),negative control group(group D),and positive control group(group E).Attenuated total reflection-Fourier transform infrared spectroscopy(ATR-FTIR),scanning electron microscopy(SEM),transmission electron microscopy(TEM),and energy-dispersive X-ray spectroscopy(EDS)were used to evaluate the remineralization effect in each group.Results ATR-FTIR showed that in group B,after remineralization for 21 d,the peak position and peak intensity in the infrared spectrum of demineralized dentin slices were approximately fitted with those of natural dentin slices.In group B,remineralization substances were deposited evenly on the surface of demineralized dentin to seal the dentin tubules,and the periodic“stripe”structures could be seen in the collagen fibers of dentin.Group B had a significantly higher calcium/phosphorus molar ratio on the surface of remineralized dentin slices than groups A,C,and D(F=19.27,P<0.05),while there was no significant difference between group B and group E(P>0.05).Conclusion CPP-ACP has an ideal bionic remineralization effect on demineralized dentin slices with the assistance of TPP.
