胃癌中长链非编码RNA SGOL1-AS1的表达变化及临床意义

摘要 目的探究长链非编码RNA(SGOL1-AS1)在胃癌中表达情况和相关临床意义。方法实时荧光定量PCR检测65例胃癌组织及癌旁组织、3株胃癌细胞系及正常胃黏膜细胞系的SGOL1-AS1表达水平,同时使用star Base在线数据库评估胃癌中SGOL1-AS1表达水平。结合临床资料分析SGOL1-AS1表达差异与临床病理特征关系,构建受试者工作曲线评估SGOL1-AS1对胃癌诊断效能,采用Kaplan-Meier法分析SGOL1-AS1表达水平与胃癌患者生存预后关系。结果SGOL1-AS1在胃癌组织中表达水平明显高于癌旁组织,胃癌细胞中SGOL1-AS1的表达水平高于胃黏膜正常细胞,与starBase在线数据库评估结果相同。SGOL1-AS1高表达与患者肿瘤浸润深度(χ^2=4.986,P=0.026)、TNM分期(χ^2=7.963,P=0.005)、肿瘤大体分型(χ^2=5.015,P=0.025)、脉管浸润(Fisher,P=0.048)均显著相关,而其与年龄、性别、吸烟、饮酒、肿瘤最大直径、淋巴结转移、远处转移、组织分化程度等均无明显相关(均P>0.05)。SGOL1-AS1检测胃癌的ROC曲线下面积为0.612(95%CI 0.514~0.710,P=0.028),灵敏度为87.70%,特异度为40.00%。SGOL1-AS1高表达组患者预后更差(Log-rank,P=0.010)。结论长链非编码RNA SGOL1-AS1在胃癌中表达明显上调,并可能调控着胃癌的发生发展过程,影响胃癌患者预后,有潜力为胃癌的诊疗开辟新的方向。 Objective To investigate the expression and related clinical significance of long-chain non-coding RNA in gastric cancer.Methods Real-time fluorescence quantitative PCR was used to detect the expression of SGOL1-AS1 in 65 cases of gastric cancer tissues and adjacent tissues,3 gastric cancer cell lines and normal gastric mucosa cell lines.At the same time,the star Base online database was used to evaluate the SGOL1-AS1 in gastric cancer.Combined with clinical data to analyze the relationship between SGOL1-AS1 expression difference and clinicopathological characteristics,a receiver operating curve was constructed to evaluate the diagnostic efficacy of SGOL1-AS1 for gastric cancer,and Kaplan-Meier method was used to analyze the relationship between SGOL1-AS1 and survival prognosis of gastric cancer patients.Results SGOL1-AS1 in gastric cancer tissue was significantly higher than that in adjacent tissues,and SGOL1-AS1 in gastric cancer cells was higher than that of normal gastric mucosa cells,which was the same as the evaluation results of star Base online database.The high expression of SGOL1-AS1 was significantly correlated with depth of tumor invasion(χ^2=4.986,P=0.026),TNM stage(χ^2=7.963,P=0.005),macroscopic types(χ^2=5.015,P=0.025),and vascular invasion(Fisher,P=0.048),but it was not significantly related to age,gender,smoking,alcohol consumption,tumor maximum diameter,lymph node metastasis,distant metastasis,and tumor differentiation(P>0.05).The area under the ROC curve of SGOL1-AS1 for detecting gastric cancer was 0.612(95%CI 0.514~0.710,P=0.028),the sensitivity was 87.70%,and the specificity was 40.00%.The prognosis of SGOL1-AS1 high expression group was worse(Log-rank,P=0.010).Conclusion Long non-coding RNA SGOL1-AS1 is significantly up-regulated in gastric cancer and may regulate the occurrence and development of gastric cancer to affect the prognosis of gastric cancer patients,which maybe open up new directions to diagnose and treat gastric cancer.
