摘要
目的探讨过表达Pannexin 3对急性牙髓炎模型大鼠牙髓组织炎症反应的作用及其机制。方法选取40只健康雄性SD大鼠,其中10只作为对照组,其余30只建立急性牙髓炎大鼠模型,随机分为模型组、抑制组、过表达组,每组10只。采用Western blot法检测核因子κB(NF-κB)、细胞外调节蛋白激酶(ERK)、p38蛋白激酶(p38MAPK)、Pannexin 3、Som、Shh、Ptc、Glil蛋白相对表达量,采用酶联免疫吸附法检测趋化因子(CX3CL1)、血红素加氧酶-1(HO-1)、血管内皮生长因子(VEGF)、白细胞介素-8(IL-8)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平。结果过表达组Pannexin 3表达高于模型组和抑制组,低于对照组(P<0.05)。过表达组大鼠NF-κB、ERK蛋白表达均低于模型组和抑制组,高于对照组;p38MAPK表达高于模型组和抑制组,低于对照组(P<0.05)。过表达组大鼠CX3CL1、HO-1、VEGF水平均低于模型组和抑制组,高于对照组(P<0.05)。过表达组大鼠IL-8、IL-6、TNF-α水平均低于模型组和抑制组,高于对照组(P<0.05)。过表达组大鼠Som、Shh、Ptc、Glil表达均低于模型组和抑制组,高于对照组(P<0.05)。结论过表达Pannexin 3能够减少急性牙髓炎模型大鼠牙髓组织炎症因子,有效抑制大鼠牙髓组织炎症反应,促进大鼠牙髓的损伤修复。
Objective To explore the effect and mechanism of overexpression of Pannexin 3 on the inflammatory reaction of pulpal tissue in rats with acute pulpitis.Methods Forty healthy male SD rats were selected,10 of which were regarded as the control group,and the remaining 30 were used to establish the acute pulpitis rat models.They were randomly divided into model group,inhibition group and overexpression group,with 10 rats in each group.The relative expressions of nuclear factorκB(NF-κB),extracellular regulatory protein kinase(ERK),p38 protein kinase(p38MAPK),Pannexin 3,Som,Shh,Ptc,Glil protein were detected by Western blot.The levels of chemokines(CX3CL1),heme oxygenase-1(HO-1),vascular endothelial growth factor(VEGF),interleukin-8(IL-8),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)were detected by enzyme-linked immunosorbent assay.Results Pannexin 3 expression in the overexpression group was higher than that in the model group and the inhibition group,while it was lower than that in the control group(P<0.05).The protein expressions of NF-κB and ERK in the overexpression group were lower than those in the model group and the inhibition group,and they were higher than those in the control group,and the expression of p38MAPK was higher than that in the model group and the inhibition group,while it was lower than that in the control group(P<0.05).The levels of CX3CL1,HO-1 and VEGF in the overexpression group were lower than those in the model group and the inhibition group,and they were higher than those in the control group(P<0.05).The levels of IL-8,IL-6 and TNF-αin the overexpression group were lower than those in the model group and the inhibition group,and they were higher than those in the control group(P<0.05).The expressions of Som,Shh,Ptc and Glil in the overexpression group were lower than those in the model group and the inhibition group,and they were higher than those in the control group(P<0.05).Conclusion Overexpression of Pannexin 3 can reduce the inflammatory factors in the pulpal tissue of acute pulpitis model rats,effectively inhibit the inflammatory reaction of rat pulpal tissue,and promote the repair of rat pulpal damage.
