摘要
目的:鉴定黄芪生脉饮中的主要天然糖类物质,并对其含量进行测定。方法:通过和标准品在带蒸发光检测器高效液相色谱(HPLC-ELSD)上出峰时间的比较,来鉴定黄芪生脉饮中的主要天然糖类成分;采用COSMOSIL 5NH2-MS氨基柱结合HPLC-ELSD进行单糖含量测定,流动相为乙腈-水(80∶20),流速为1 mL/min,柱温35℃。结果:黄芪生脉饮中的主要天然糖类成分为果糖;基于氨基柱建立的HPLC-ELSD果糖检测方法在0.2~16 mg/mL的浓度范围内线性关系良好,平均回收率为98.63%,RSD为1.76%。结论:黄芪生脉饮中含有较高的天然果糖成分,其含量可用HPLC-ELSD法进行检测。
Objective: To identify the main natural saccharides in Huangqi Shengmai yin, and to determinate their contents. Methods: The main natural saccharides in Huangqi Shengmai yin were identified by comparing the peak time on high-performance liquid chromatography-evaporative light scattering detector(HPLC-ELSD) with that of standard reference substance. The content determination of monosaccharide was performed by COSMOSIL 5 NH2-MS amino column combined with HPLC-ELSD;the mobile phase was acetonitrile-water(80:20);the flow rate was 1 mL/min;the column temperature was 35 ℃. Results:The main natural saccharide in Huangqi Shengmai yin was fructose. The HPLC-ELSD fructose detection method based on amino column showed good linear relationship in the concentration range of 0.2 to 16 mg/mL, with an average recovery rate of 98.63% and RSD of 1.76%. Conclusion: Huangqi Shengmai yin has relatively high content of fructose which can be detected via HPLC-ELSD method.
