摘要
内质网应激(Endoplasmic reticulum stress,ERS)与许多代谢性疾病有关,比如胰岛素抵抗、脂肪肝等营养代谢性疾病。脂肪肝是围产期奶牛常见多发的营养代谢性疾病,严重危害着奶牛养殖业的发展。含黄素单加氧酶3(Flavin containing monooxygenase,FMO3)是近年来发现的与内质网应激有密切联系的调节分子。因此,我们推测FMO3有可能参与调节奶牛的内质网应激过程,从而影响脂质代谢。试验通过体外培养犊牛原代肝细胞感染FMO3过表达和低表达腺病毒载体,检测FMO3过表达和低表达对NEFA介导的犊牛原代肝细胞内质网应激的影响,以明确FMO3对奶牛肝细胞内质网应激的调节作用。结果发现,FMO3过表达腺病毒载体明显增加了NEFA介导的奶牛肝细胞内质网应激相关因子的mRNA表达水平,低表达腺病毒明显降低了NEFA介导的内质网应激相关因子的mRNA表达水平,这为阐明奶牛内质网应激的发病机制提供了一定的理论依据。
Endoplasmic reticulum stress is associated with many metabolic diseases,such as insulin resistance,fatty liver and other nutritional and metabolic diseases.Fatty liver is a common nutritional and metabolic disease in perinatal dairy cows,which seriously endangers the development of dairy industry.Flavin containing monooxygenase3(FMO3)was a regulatory molecule,which is closely related to endoplasmic reticulum stress.Therefore,we speculate that FMO3 may be involved in the regulation of endoplasmic reticulum stress in dairy cows,thus affecting lipid metabolism.In order to determine the regulatory effect of FMO3 on endoplasmic reticulum stress in bovine hepatocytes,primary calf hepatocytes were cultured in vitro and infected with FMO3 overexpression and low expression adenoviral vectors to detect the effects of FMO3 on NEFA-mediated endoplasmic reticulum stress in calf primary hepatocytes.The results showed that FMO3 overexpression adenovirus vector significantly increased the mRNA expression of endoplasmic reticulum stress related factors mediated by NEFA,while low expression adenovirus significantly decreased the mRNA expression level of endoplasmic reticulum stress related factors mediated by NEFA,which provided a theoretical basis for elucidating the pathogenesis of endoplasmic reticulum stress in dairy cows.
