摘要
将猪细小病毒L株原始种毒E0代,在ST传代细胞上连续传代至E12代,检测E1~E12代病毒液的病毒含量和红细胞凝集价,并取E1、E3、E5、E7、E9和E12代病毒液进行了毒力、免疫原性、特异性和纯净性检验,来确定该病毒的遗传稳定性。试验结果表明,E1~E12代病毒液,病毒含量为107.36~107.60TCID50/mL、红细胞凝集价在1512~11024;E1、E3、E5、E7、E9和E12代病毒液,毒力稳定、免疫原性良好、特异、纯净。为进一步确保L株作为生产用毒种的遗传稳定性,将其作为生产用毒种的基础代次限定为E1~E5代,生产代次限定为E6~E8代,生产毒种最高使用代次不超过3代。
L Strain of Porcine Parvovirus in the original kind of poison,EO generation in ST consecutive batches to E12 generation to extend cell generation,detection of E1-E12 generation of the concentrations of the virus disease venom and erythrocyte aggregation,and E1,E3,E5,E7,E9 and E12 generation disease venom,the virulence,immunogenicity,specificity and pure test,to determine the genetic stability of the virus.The results showed that the virus content of E1-E12 generation virus was 107.36-107.60 TCID/mL and the erythrocyte agglutination value was 1:512-1:1024.E1,E3,E5,E7,E9 and E12 generations of virus venom,stable virulence,good immunogenicity,specificity,purity.To ensure the genetic stability of Strain L as a production strain,the basic generation of strain L as a production strain was limited to E1-E5 generation and the production generation was limited to E6-E8 generation,and the maximum use generation of the production strain was no more than 3 generation.
