木糖醇饮料对溃疡性结肠炎小鼠结肠病变的影响 被引量:1

摘要 目的考察木糖醇饮料对溃疡性结肠炎小鼠结肠病变的影响。方法SPF雄性小鼠随机分为正常组、溃疡性结肠炎(ulcerative colitis,UC)模型组及木糖醇饮料高、中、低剂量组。正常组给予蒸馏水;模型组给予2.5%硫酸葡聚糖钠(DSS)溶液自由饮用诱导其溃疡性结肠炎;木糖醇饮料高、中、低剂量组分别给予含有不同浓度木糖醇(10%、5%、2.5%)的DSS溶液。各组小鼠连续自由饮用相应液体10 d。实验结束时,检测各组小鼠结肠的病理变化,同时测定结肠紧密连接蛋白(ZO-1与Occludin)、钠葡萄糖转运蛋白(SGLT-1)和钠氢协同转运蛋白(NHE3)的表达水平。结果与正常组比较,DSS造模后的UC小鼠出现稀便,疾病活动指数显著升高(P<0.05)。与UC模型组比较,木糖醇饮料组肠道长度显著缩短、肿胀,结肠黏连和坏死的情况明显加重,ZO-1、Occludi、SGLT-1及NHE3的表达显著下降(P<0.05或P<0.01)。结论高浓度木糖醇饮料可加剧UC小鼠结肠病变及屏障功能的破坏。 Objective To investigate the hazard effects of xylitol beverage on the permeability of colonic epithelial barrier in colitis mice.Methods Male Kunming mice were randomly divided to five groups.Animals in the normal group were supplied with distilled water and the others were received 2.5%DSS daily.DSS+xylitol groups were given 2.5%DSS solution and 2.5%,5%or 10%xylitol for 10 days.The pathological changes of colon tissue were examined.The expression of ZO-1,Occludin,SGLT-1 and NHE3 was detected.Results Compared with the normal group,the UC mice induced by DSS had loose stools and the disease activity index increased significantly(P<0.05).Compared with UC control mice,the length of intestine in xylitol beverage group was significantly shortened and swollen,the adhesion and necrosis of colon were significantly increased,and the expressions of ZO-1,occludi,SGLT-1 and NHE3 were significantly decreased(P<0.05,P<0.01).Conclusion High concentration xylitol beverage can aggravate colonic lesion and damage of barrier function in UC mice.
