摘要 目的建立高效液相色谱法(HPLC)同时测定膜苞鸢尾中两种有效黄酮类成分野鸢尾苷和野鸢尾黄素含量。方法色谱柱为VenusilASB C18(250mm×4.6mm,5μm),流动相为乙腈-0.5%H3PO4水溶液,流速1.0mL·min-1,检测波长269nm,柱温25℃,进样量5μl。结果野鸢尾苷和野鸢尾黄素质量浓度分别为2.4~12μg·mL-1(r=0.9996)、2.24~11.2μg·mL-1(r=0.9995)的范围内与峰面积呈良好的线性关系;平均加样回收率及相应RSD分别为98.45%(RSD=2.29%)、97.93%(RSD=3.93%)(n=6)。结论该方法准确度好,精密度高,本实验可为膜苞鸢尾的开发利用和质量控制提供实验依据。 OBJECTIVE To establish an HPLC method for the simultaneous determination of twoeffective flavonoids in Irisscariosa.METHODS The method was performed on a column of VenusilASB C18(250mm×4.6mm,5μm)with the mobile phase consisting of acetonitrile-0.5%phosphoric acid solution with gradient elution at the flow rate of 1.0ml·min-1,the detection wavelength was 269nm,the column temperature was 25℃and the sample size was 5μL.RESULTS The linear ranges of iridin and irigeninwere 2.4~12μg·mL-1(r=0.9996)、2.24~11.2μg·mL-1(r=0.9995),respectively.The average recovery and the corresponding RSD were 98.45%(RSD=2.29%)、97.93%(RSD=3.93%)(n=6),respectively.CONCLUSION The method is accurate with high precision,which can provide experimental basis for the development,utilization and quality control ofIrisscariosa.
