摘要
目的通过远志薄层色谱及生物自显影色谱分析,为远志质量评价及对抑制乙酰胆碱酯酶活性成分筛选提供支持。方法采用薄层色谱法分离和自显影活性测定技术,对不同来源远志进行多成分分离及抑制乙酰胆碱酯酶活性分析。结果远志药材可见十二个薄层色谱主斑点,远志汕酮Ⅲ、3,6’-二芥子酰基蔗糖、远志糖苷A可用于远志药材鉴别;细叶远志皂苷、远志皂苷元可用于远志药材质量评价。利用生物自显影色谱斑点初步筛选出1个活性成分。结论远志薄层色谱鉴别方法简便易行,特征性强,可以用于不同远志药材鉴别以及评价药材质量优劣;薄层生物自显影色谱适用于远志乙酰胆碱酯酶抑制成分的筛选。
OBJECTIVE To provid supports for quality evaluation and active ingredients screening of Polygala Radix by TLC and auto-chromatographic chromatography. METHODS Adopt TLC separation and auto-development activity measurement technology, Multi-component separation and analysis of inhibition of acetylcholinesterase activity from Polygala from different sources. RESULTS There are twelve main spots of TLC in Polygala medicinal materials. Polygalaxanthone Ⅲ,3,6’-disinapoyl sucrose and tenuifoliside A can be used for identification of Polygala medicinal materials.Tenuifolin and senegenin can be used for polygala quality evaluation.An active ingredient was preliminarily screened by using auto-chromatographic spots. CONCLUSION Polygala TLC identification method is simple and easy, with strong characteristics. TLC can be used to identify different Polygala medicinal materials and evaluate the quality of medicinal materials. TLC-bioautography is suitable for screening acetylcholinesterase inhibitory ingredients from Polygala Radix.
