摘要
为揭示鸭肠炎病毒(DEV)感染对鸭肠道菌群多样性的影响,本研究采用16S rRNA基因高通量测序和Illumina Hiseq 2500高通量测序技术对DEV感染后鸭肠道菌群多样性和丰度进行分析。结果显示:与对照组比较,感染组鸭肠道菌群Alpha多样性指数均呈先升高后降低趋势,尤其是在DEV感染96 h和120 h后,鸭肠道菌群Beta多样性(Weighted Unifrac指数)距离越远,相似度越低;DEV感染后鸭肠道优势菌群主要分布于厚壁菌门、变形菌门、拟杆菌门和梭杆菌门,梭菌纲、丙型变形菌纲、拟杆菌纲和梭杆菌纲,梭菌目、肠杆菌目、拟杆菌目和梭杆菌目,肠杆菌科、消化链球菌科、拟杆菌科和梭杆菌科,大肠杆菌志贺菌亚属、拟杆菌属、梭菌属和Turicibacter属,尤其是产气荚膜梭菌和坏死梭杆菌呈先下降后升高趋势,这可能与DEV感染所导致的肠道病变存在一定的关联性。本研究结果为DEV致病机理及其所致疾病的防控研究奠定了基础。
To determine the effects of duck enteritis virus(DEV) infection on the diversity of intestinal flora in ducks, the highthroughput sequencing of bacterial 16 S ribosomal RNA(rRNA) gene on Illumina Hiseq 2500 platform was performed, and the diversity and abundance of intestinal flora in ducks infected with DEV were analyzed. The results showed that, upon DEV infection,the Alpha diversity index of intestinal flora in the infection group was increased at the early time post infection but decreased later.The analysis of beta diversity(Weighted unifrac index) of duck intestinal flora suggested lower similarity between the infected ducks and control ducks at 96 and 120 hours post DEV infection. The dominance of duck intestinal flora after DEV infection was mainly distributed in Phytophthora, Proteus, Bacteroides and Clostridium. It was noteworthy that the abundance of Clostridium perfringens and Clostridium necrosis decreased at first and increased subsequently after DEV infection, which might be associated with the intestinal lesions caused by infection. These results provide a scientific basis for further illustration of the pathogenic mechanism of DEV infection and the prevention and control of the disease caused by this virus.
