基于网络药理学探讨参附注射液中人参皂苷及生物碱治疗病态窦房结综合征的作用机制 被引量:4

摘要 目的利用网络药理学方法探讨参附注射液(SFI)中人参皂苷及生物碱治疗病态窦房结综合征(SSS)的作用机制。方法通过文献检索获取SFI中人参皂苷及生物碱的成分数据,利用SEA、Hit Pick、Swiss Target Prediction数据库查找成分预测靶点,与从Gene Cards中获得的疾病靶点进行比对后构建成分-映射靶点网络及蛋白互作网络,利用DAVID对映射靶点进行KEGG通路富集分析。结果本研究共搜集到24个人参皂苷、22个生物碱成分和300个成分靶点,其中47个靶点与SSS相关。KEGG通路主要涉及细胞凋亡、炎症反应、组织修复、调节离子通道功能等。结论SFI治疗SSS的过程涉及多个成分、多个靶点和多条通路,主要与调节离子通道功能、组织修复、抑制细胞凋亡和炎症反应等机制密切相关。 Objective:To explore the mechanism of ginsenoside and alkaloid in Shenfu Injection(SFI)on sick sinus syndrome(SSS)based on network pharmacology.Methods:Literature mining was used to obtain the compounds data of ginsenoside and alkaloid in SFI.SEA,HitPick and Swiss Target Prediction databases were used to find the prediction targets of components,and then compared with disease targets screened by Gene Cards database.The component-mapping target network and protein interaction network were constructed by Cytoscape software.KEGG pathway enrichment analysis of the mapping targets were performed with DAVID database.Results:Totally 24 ginsenosides,22 alkaloids and 300 component targets were collected,47 of which were related to SSS.KEGG pathway was mainly involved in apoptosis,inflammatory response,tissue repair,regulation of ion channel function and so on.Conclusion:The treatment of SSS with SFI involves many components,targets and pathways,closely related to the mechanisms of regulating ion channel function,repairing tissue,inhibiting apoptosis and responding inflammatory.
