益气和胃止痛方对急性胃炎模型大鼠胃黏膜NO、IL-1和G17表达的影响 被引量:3

Effect of Yiqi Hewei Zhitong Recipe on the Expression of NO,IL-1 and G17 in Gastric Mucosa of Rats with Acute Gastritis and Its Mechanism

摘要 目的探讨益气和胃止痛方对急性胃炎模型大鼠胃黏膜一氧化氮(NO)、白细胞介素-1(IL-1)、胃泌素-17(G17)表达的影响及作用机制。方法将60只SD大鼠随机分为对照组、模型组、阳性组和中药低剂量组、中剂量组和高剂量组,每组10只。模型组、中药组和阳性组采用灌胃法制备急性胃炎大鼠模型,对照组和模型组给予生理盐水灌胃,阳性组给予法莫替丁灌胃,中药组给予益气和胃止痛方灌胃。硝酸还原酶法测定胃黏膜NO含量,ELISA法检测胃黏膜IL-1水平以及丙二醛(MDA)含量,免疫法检测血清胃蛋白酶原(PGR)以及G17表达水平。结果与对照组比较,模型组胃黏膜表面上模型组大鼠的胃黏膜萎缩脱落或缺损,上皮出血水肿,腺体排列紊乱且明显减少,腺体囊性扩张,可见中性粒细胞浸润;大鼠的胃黏膜中IL-1、MDA水平及PGR水平明显升高,胃黏膜中NO及G17表达明显降低(P<0.05)。给药后,与模型组比较,阳性组和低剂量组大鼠的胃黏膜萎缩程度明显减轻,腺体明显增多,排列稍紊乱,固有层内可见小血管充血及中性粒细胞浸润,大鼠的溃疡指数明显降低,溃疡抑制率明显升高,大鼠的胃黏膜中IL-1、MDA水平及PGR水平明显降低,胃黏膜中NO以及G17表达明显升高(P<0.05),且阳性组和高剂量组间比较各指标具有显著性差异(P<0.05)。结论益气和胃止痛方可抑制急性胃炎模型大鼠胃黏膜的溃疡,提高胃黏膜NO含量,降低IL-1、PGR和MDA水平,调整胃黏膜中的G17表达,其作用机制可能与调节NO、IL-1水平和G17表达有关。 Objective:To investigate the effect of Yiqi Hewei Zhitong Recipe on the expression of NO,IL-1 and G17 in gastric mucosa of rats with acute gastritis and its mechanism.Methods:A total of 60 SD rats were randomly divided into control group,model group,positive group and test group(low-dose group,middle-dose group and high-dose group),10 rats in each group.The model group,the test group and the positive group were prepared by gastrointestinal administration.Rats in the control group and the model group were administered with saline,the positive group was administered with famotidine by gavage,and the test group was given Yiqi Hewei Zhitong Recipe.The nitrate reductase method was used to determine the NO content in the gastric mucosa,the Elisa method was used to detect the gastric mucosal interleukin-1(IL-1)level and MDA content,and the immunological method was used to detect the serum pepsinogen(PGR)and G17 expression levels.Results:Compared with the control group,the gastric mucosa of the model group had atrophy or loss of gastric mucosa,epithelial blood edema,disordered and significantly reduced glands,cystic expansion of the glands,and infiltration of neutrophils.The levels of IL-1,MDA and PGR in the gastric mucosa of rats were significantly increased,and the levels of NO and G17 in the gastric mucosa were significantly decreased(P<0.05).After administration,compared with the model group,the gastric mucosal atrophy of the rats in the positive group and the low-dose test group was significantly reduced,the glands were significantly increased,and the arrangement was slightly disordered.Small blood vessel congestion and neutrophil infiltration were seen in the lamina propria.The ulcer index of the rats was significantly re duced,and the ulcer inhibition rate was significantly increased.The levels of IL-1,MDA and PGR in the gastric mucosa of rats were significantly reduced,and the levels of NO and G17 in the gastric mucosa were significantly increased(P<0.05);there was significant difference between the positive group and the high-dose test group(P<0.05).Conclusion:Yiqi Hewei Zhitong Recipe can inhibit gastric mucosa ulcer in rats with acute gastritis model,increase NO content and PGR level in gastric mucosa,decrease IL-1 and MDA levels,and adjust G17 expression in gastric mucosa.Its mechanism may be related to the levels of NO and IL-1,as well as the expression of G17.
