抑制miR-621表达促进人胃癌细胞系增殖、迁移和侵袭

Inhibition of miR-621 expression promotes proliferation,migration and invasion of human gastric cancer cell lines

目的检测miR-621在胃癌组织及胃癌细胞系中的表达情况,评估miR-621的预后价值,分析miR-621对胃癌细胞生物学功能的影响。方法荧光定量PCR检测癌和癌旁组织以及胃癌细胞系中miR-621的表达。用卡方检验分析miR-621与胃癌患者临床病理学特征之间的关系;用生存曲线和Cox回归分析miR-621对胃癌的预后价值。最后转染胃癌细胞miR-621的模拟物和抑制剂,通过CCK-8法、Transwell小室法分析胃癌细胞增殖、迁移和侵袭能力。结果胃癌组织以及胃癌细胞系中miR-621的表达量显著低于对照组(P<0.01),且与胃癌患者的淋巴结转移、肿瘤组织分化和TNM分期显著相关(P<0.05)。同时发现miR-621可以作为胃癌患者的独立预后因子(HR=3.09,95%CI=1.31~7.27,P=0.01),且miR-621表达水平低的患者生存率较差(log rank test P<0.001)。此外,抑制miR-621的表达水平能显著的增加胃癌细胞的增殖、迁移和侵袭的能力(P<0.05)。结论miR-621的低表达与胃癌的不良预后相关,并且会促进胃癌细胞的增殖、迁移和侵袭能力。

Objective To detect the expression of miR-621 in gastric cancer tissues and cell lines and to evaluate the prognostic significance of miR-621,and to analyze the effect of miR-621 on the biological function of gastric cancer cells.Methods Expression of miR-621 in cancer tissue and cell lines was detected by real-time polymorperase chain reaction(RT-qPCR).Chi-square test was used to analyze the relationship between miR-621 and clinical pathological features of gastric cancer patients.Prognostic significance of miR-621 for gastric cancer was analyzed by survival curve and Cox regression.Finally,human gastric cancer cells were transfected with miR-621 mimics and inhibitors to analyze the proliferation,migration and invasion ability of gastric cancer cells.ResultsThe expression of miR-621 was significantly downregulated in gastric cancer tissues and cell lines as compared to the adjacent tissues and gastric mucosa cell lines(P<0.01),and low expression was associated with lymph node metastasis,tumor differentiation,and TNM stage in gastric cancer(P<0.05).The patients with low expression of miR-621 had a poor overall survival rates(log-rank test P<0.001),and miR-621 was an independent prognostic indicator for gastric cancer patients(HR=3.09,95%CI=1.31-7.27,P=0.01).In addition,low-expression ofmiR-621 significantly increased proliferation,migration,and invasion of gastric cancer cells(P<0.05).Conclusions Low expression of miR-621 is associated with poor prognosis of gastric cancer,which promotes the proliferation,migration and invasion of gastric cancer cells.
