摘要
目的:观察电针对2型糖尿病ZDF大鼠血糖、PI3K p85及GLUT4蛋白表达影响。方法:选取14只体质量140~160 g(2月龄)SPF级雄性ZDF(fa/fa)大鼠,随机分为模型组、电针组;另取7只同月龄SPF级雄性(fa/+)大鼠作为对照组。连续干预4周,测定各组空腹血糖。4周后Western blot检测骨骼肌的磷脂酰肌醇3激酶(PI3K)和葡萄糖转运蛋白4(GLUT4)蛋白表达水平。结果:干预后与对照组比较,模型组空腹血糖显著升高(P<0.01);与模型组比较,电针组空腹血糖显著下降(P<0.05)。Western bolt检测,与对照组对比,模型组PI3K p85、GLUT4蛋白表达量显著下降(P<0.01);与模型组比较,电针组PI3K p85、GLUT4蛋白表达显著升高(P<0.01)。结论:电针干预可改善ZDF大鼠空腹血糖及骨骼肌细胞的胰岛素抵抗(IR)。其机制可能与PI3K-GLUT4信号传导途径有关。
Objective:To observe the effects of electro-acupuncture on blood sugar and the protein expressions of PI3Kp85 and GLUT4 in Zucker Diabetic Fatty(ZDF)rats with type 2 diabetes mellitus(DM).Methods:14 SPF male ZDF(fa/fa)rats weighing 140 g^160 g were randomly divided into the model group and the electro-acupuncture group,and another 7 SPF male Zucker Lean,ZL(fa/+)rats were taken as the control.After 4 weeks of intervention,fasting blood glucose was measured in each group.Western blot method was used to detect the protein expressions of phosphatidylinositol 3-kinase(PI3K)and glucose transporter 4(GLUT4)in skeletal muscle.Results:The fasting blood sugar in the model group increased significantly compared to that in the model group(P<0.01);after electro-acupuncture intervention,it decreased significantly(P<0.05).Compared to the control group,the protein expression of PI3K p85 and GLUT4 decreased significantly in the model group(P<0.01);after electro-acupuncture intervention,they increased significantly(P<0.01).Conclusion:Electro-acupuncture can improve fasting blood sugar and insulin resistance(IR)of skeletal muscle cells in ZDF rats.The mechanism may be related to regulating PI3K-GLUT4 signal pathway.
