Expression and clinical significance of Hedgehog signaling pathway related proteins in head and neck squamous cell carcinoma

摘要 目的:探讨头颈鳞状细胞癌(head and neck squamous cell carcinoma,HNSCC)中Hedgehog信号通路成员Shh、Ptch1和Gli1蛋白表达的关系及其意义。方法:利用生物信息学方法分析Shh、Ptch1、Gli1在HNSCC和正常组织中的表达差异预测本研究可行性,采用免疫组化法检测84例头颈鳞状细胞癌及10例正常黏膜上皮组织中Shh、Ptch1和Gli1蛋白表达,分析Shh、Ptch1和Gli1蛋白表达水平与头颈鳞状细胞癌患者临床病理特征的关系。结果:头颈鳞状细胞癌组织中Shh、Ptch1和Gli1蛋白呈高表达(分别为65.5%、60.7%和63.1%),均高于正常黏膜上皮组织(分别为20%、30%和30%)。Shh蛋白表达与头颈鳞状细胞癌患者临床分期和淋巴结转移有关(P<0.05),而与患者年龄、性别、肿瘤局部侵袭和远处转移均无关(P>0.05);Ptch1蛋白表达与头颈鳞状细胞癌患者临床病理特征均无关(P>0.05);Gli1蛋白表达仅与头颈鳞状细胞癌患者淋巴结转移有关(P<0.05)。Shh、Ptch1和Gli1蛋白的表达水平在鼻咽鳞癌和口咽鳞癌中没有差异。Shh与Ptch1、Gli1蛋白表达呈正相关。结论:Shh、Ptch1和Gli1蛋白在头颈鳞状细胞癌中高表达,与头颈鳞状细胞癌的发生、发展密切相关,是头颈鳞状细胞癌防治的一个重要靶标。 Objective:To investigate the relationship and significance of Shh,Ptch1 and Gli1 protein expression of Hedgehog signaling pathway in head and neck squamous cell carcinoma(HNSCC).Methods:Bioinformatics method was used to analyze the feasibility of predicting the differential expression of Shh,Ptch1 and Gli1 in HNSCC and normal tissues.Immunohistochemical method was used to detect the expression of Shh,Ptch1 and Gli1 in 84 cases of HNSCC and 10 cases of normal mucosal epithelium.The relationship between the expression of Shh,Ptch1 and Gli1 protein and the clinical pathological features of HNSCC patients was analyzed.Results:The expression of Shh,Ptch1 and Gli1 proteins in HNSCC was 65.5%,60.7% and 63.1%,respectively,which was higher than that in normal mucosal epithelium,which was 20%,30% and 30%,respectively.The expression of Shh protein was related to clinical stage and lymph node metastasis of HNSCC patients,but not to age,gender,local invasion and distant metastasis of the patients.There was no correlation between the expression of Ptch1 protein and the clinical pathological features of HNSCC patients.The expression of Gli1 protein was only related to lymph node metastasis of HNSCC patients.There was no significant difference in the expression of Shh,Ptch1 and Gli1 protein between nasopharyngeal squamous cell carcinoma and oropharyngeal squamous cell carcinoma.Shh was positively correlated with Ptch1 and Gli1 protein expression.Conclusion:The expression of Shh,Ptch1 and Gli1 proteins was high in HNSCC and is related with development of HNSCC,which is a important target of HNSCC prevention.
