摘要
为了比较三亚西瑁岛优势种花环肉芝软珊瑚(Sarcophyton ehrenbergi)与短指软珊瑚(Sinularia ceramensis Verseveldt)转录组的数据,以高通量测序技术对2种软珊瑚进行转录组测序分析.利用Trinity软件对所有reads从头组装后,得到130587条单基因簇(Unigenes).通过Blast比对SwissProt和Hmmscan搜索Pfam蛋白同源序列,共得到86002条编码蛋白框(CDS).与Nr(Non-redundant,非冗余)、KOG(Clusters of orthologous groups for eukaryotic complete genomes,真核生物蛋白相邻类的聚簇)、KEGG(Kyoto encyclopedia of genes and genomes,京都基因与基因组百科全书)、Swissprot 4大数据库比对,共获得74970条注释基因,占总Unigene数的57.41%.2种不同珊瑚在相同环境中基因表达存在差异,短指珊瑚冠部Unigene数目远大于其茎部基因数,差异明显.花环肉芝软珊瑚GC含量都略大于短指珊瑚.在KEGG功能分类比对中,花环肉芝软珊瑚与短指珊瑚转录组中信号传导功能基因包含的Unigene数目最多,为8232条,包含次级代谢产物生物合成功能的Unigene数最少,仅有45条.本研究丰富了南海软珊瑚转录组数据库.
The purpose of this study is to obtain the transcriptome data of Sarcophyton and Sinularia soft corals in Ximao Island,Sanya,by using high-throughput transcriptome sequencing technology.130,587 unigenes were obtained from the two species after all reads were reassembled by Trinity software.By Blast comparison between Swissprot and Hmmscan and by searching for Pfam protein homologous sequences,86,002 of coding protein frames(CDS)were obtained.By comparing those CDS with the four databases of Nr,KOG,KEGG and Swissport,obtained were 74,970 annotation genes,accounting for 57.41%of the total number of unigenes.There are differences in gene expression between the two soft corals species under the same environment.The number of transcriptome UniGene from the crown of Sinularia body is much larger than that of its stem.The GC content of Sarcophyton was slightly higher than that of Sinularia.Through comparison of functional classification in the KEGG,the number of unigenes of signaling function in two soft corals transcriptome were maximum,that is,8,232,and the number of unigenes of the biosynthesis of secondary metabolites was minimum,that is,45.The current research enriches the transcriptome database of soft corals.
