摘要
从三亚近海海水中分离出一株海洋发光细菌,命名为L2,并对其进行了生化及分子鉴定,初步确定该菌株为发光杆菌属(Photobacterium)的鳆鱼发光杆菌(P.leiognathi).发光特性分析结果表明:该菌在20~35℃的温度范围内均能发光,其中,25~30℃发光强度较高;该菌株具有较广的耐受盐能力,可以在1%~7%的氯化钠浓度范围发光,其中,在4%的氯化钠浓度下发光能力最高;此外,该菌还具有较强的pH适应范围,可在pH值为5~10的范围内发光,pH低于5或高于10,发光特性迅速消失.本研究结果为发光细菌L2应用于潟湖、海洋、盐碱地等环境进行污染物检测提供了一定的理论依据.
A luminescent bacterium,L2,was isolated from Sanya bay,China.Strain L2 is highly similar to Photobacterium leiognathi in terms of the biochemical characteristics and based on 16S rRNA sequence analysis.According to the analysis of luminescent characteristics,it was found that the strain luminesces in 20-35℃,with a higher luminous intensity in 25-30℃.The strain also has a wide tolerance for the concentration of sodium chloride,and it luminesces in 1%-7%sodium chloride concentration,with the strongest luminous intensity of L2 at 4%sodium chloride concentration.In addition,this strain has a tolerance for a wide range of pH.It luminesces from pH 5 to pH 10.However,the luminescence characteristics of L2 immediately disappears when pH≤5 or pH≥10.The aforementioned results provide a theoretical basis for the application of luminous bacteria L2 in detecting the environmental pollutants in lagoon,ocean,saline alkali land and other environments.
