益气温经法对OVX大鼠骨密度和骨转换标志物的影响 被引量:1

Effect of Benefiting Qi and Warming Channel Method on Bone Mineral Density and Serum Bone Turnover Markers in Ovariectomized Rats

摘要 目的探讨益气温经的地仙壮骨方对OVX大鼠骨密度和骨转换标志物的影响。方法将40只6月龄SD雌性大鼠分为:假手术组、模型组、地仙壮骨方组、阿仑膦酸钠组4组,每组10只。经去势手术2周后,阿仑膦酸钠组每周每只喂服阿仑膦酸钠混悬液2 mL(8.2 mg/kg),地仙壮骨方组每天每只喂服地仙壮骨方浓缩液2 mL(9.6 g/kg),假手术组与模型组予等量生理盐水。实验结束后,采用双能X线骨密度测量仪测量大鼠左侧股骨骨密度,ELISA法检测大鼠血清骨转换标志物:骨形成标志物P1NP和骨吸收标志物S-CTX。结果地仙壮骨方干预3个月后OVX大鼠血清中骨吸收标志物S-CTX的表达水平下降;骨形成标志物P1NP水平升高。地仙壮骨方干预后OVX小鼠股骨的骨密度增加。结论地仙壮骨方能够抑制OVX大鼠的骨量丢失和骨强度下降,改善OVX大鼠的骨代谢,可提高OVX大鼠的骨密度。 Objective To investigate the effects of Dixian Zhuanggu compound on bone mineral density(BMD)and bone turnover markers(BTMs)in ovariectomized(OVX)rats.Methods Ten 6-month old SD rats were randomly selected as sham operation group,another 30 rats were selected to establish ovariectomized models.In 2 weeks after the operation,the successful models of 30 rats were divided into model group,Dexian Zhuanggu compound group,alendronate group,each group had 10 rats.The sham operation group and model group was given the same volume of normal saline by intragastric administration,Dexian Zhuanggu compound group was treated with 2ml(9.6g/kg)by administration once per day and alendronate group 2ml(8.2g/kg)by administration once per week and for 3 months.At the end of the intervention,bone mineral density of the left femur in rats was measured by dual energy X-ray absorptiontry;the levels of bone formation markers procollagen type 1 N-peptide(P1NP)and bone resorption markers serum C-terminal telpeptide of type 1 collagen(S-CTX)in the serum of rats were detected by ELISA kit.Results After 3 months of Dexian Zhuanggu compound intervention,the expression of bone resorption markers S-CTX decreased,but the levels of bone formation markers P1NP increased.After Dexian Zhuanggu compound intervention,bone mineral density of the thigh bone increased in OVX mice.Conclusion Dexian Zhuanggu compound could exert anti-osteoporosis effect by increasing bone mineral density and strength through regulation of bone metabolism and mineral content in OVX rats.
