慢性萎缩性胃炎痰瘀互结证患者胃窦组织中Lgr5、E-cad、N-cad、p65蛋白及miR-21表达研究

Expressions of Lgr5,E-cad,N-cad,NF-κB,and miR-21 in the Gastric Antrum Tissues of Chronic Atrophic Gastritis Patients with Accumulation of Phlegm and Blood Stasis Syndrome

目的观察慢性萎缩性胃炎痰瘀互结证患者胃窦组织中Lgr5、E-cad、N-cad、p65蛋白及miR-21表达变化。方法33例慢性萎缩性胃炎痰瘀互结证患者(研究组)作为研究对象,33名健康志愿者作为健康对照组(对照组)。电子胃镜组织活检获取两组胃窦黏膜组织,采用Western Blot技术观察两组胃窦黏膜组织中Lgr5、E-cad、N-cad、p65蛋白表达的差异;采用实时荧光定量PCR检测两组胃窦黏膜组织中miR-21表达变化。结果研究组Lgr5、N-cad、p65蛋白相对表达量较对照组升高(P<0.01),E-cad蛋白相对表达量较对照组降低(P<0.01);研究组miR-21相对表达量较对照组升高(P<0.01)。结论慢性萎缩性胃炎痰瘀互结证患者胃窦组织中Lgr5、E-cad、N-cad、p65蛋白及miR-21表达异常,可能与慢性萎缩性胃炎的发生有关。

To observe the expressions of Lgr5 protein,E-cad protein,N-cad protein,p65 protein(NF-κB),and miR-21 in gastric antrum tissues of chronic atrophic gastritis(CAG)patients with accumulation of phlegm and blood stasis syndrome(APBSS).Methods Totally 33 CAG patients with APBSS were recruited as the study group,while 33 healthy volunteers were recruited as the healthy control group(Control group).Their gastric antrum tissues were collected by biopsy under electronic gastroscope.The expressions of Lgr5 protein,E-cad protein,N-cad protein,and p65 protein(NF-κB)in different gastric antrum tissues were observed by Western Blot,and the expressions of miR-21 in different gastric antrum tissues were detected by real-time fluorescence quantitative PCR.Results The relative expressions of Lgr5,N-cad protein,and p65 protein(NF-κB)in the study group were higher than those in the control group(P<0.01),and the relative expressions of E-cad protein in the study group were lower than those in the control group(P<0.01).The relative expression level of miR-21 in the study group was higher than that in the control group(P<0.01).Conclusion The expressions of Lgr5 protein,E-cad protein,N-cad protein,p65 protein(NF-κB),and miR-21 in the gastric antrum tissues of CAG patients with APBSS were related to the occurrence of CAG.
