下调NRSF的表达可通过海马通路抑制卵巢癌细胞的增殖

摘要 目的:研究神经元抑制性沉默因子(NRSF)的表达水平与卵巢癌的相关性。方法:NRSF在正常卵巢和卵巢癌细胞中的表达是通过定量PCR,western blotting法以及免疫组化等方法检测的。免疫荧光法等可以用来研究NRSF影响卵巢癌细胞增殖的机制。结果:研究结果表明NRSF在卵巢癌细胞中的表达水平显著升高,下调NRSF表达水平可以抑制卵巢癌细胞的增殖,且其表达水平与卵巢癌病人的生存时间成反比。NRSF还能通过调节海马通路(hippo pathway)的基因转录来影响卵巢癌细胞增殖。结论:我们的研究表明NRSF与卵巢癌的进展有关,NRSF可能会成为一个早期诊断卵巢癌的标志物,并且抑制NRSF的表达可能会成为治疗卵巢癌的有效方法。 Objective:This study aimed to determine the relationship between expression of NRSF and ovarian cancer.Methods:Expression of NRSF in normal ovary and ovarian cancer cells were tested by PCR(qPCR).NRSF expression in normal ovary and ovarian cancer tissue samples were examined by qPCR,western blotting and immunohistochemistry(IHC).Colony formation,anchorage-independent growth assay were applied to examine the effect of NRSF on ovarian cancer cell proliferation.Results:The results demonstrated that NRSF is significantly upregulated in ovarian cancer cells and tissues and negatively related with the survival of patients with ovarian cancer,and decrease of NRSF inhibit proliferation of ovarian cancer cells.Further analysis showed that NRSF can affect G1/S transition of cell cycle by activiting the transcription of Hippo pathway.Conclusion:Our study suggests that NRSF is related with the progression of ovarian cancer,and NRSF may be a useful early detection marker of ovarian cancer and inhibiting NRSF expression may be an good way to treat ovarian cancer.
