茯苓对去卵巢大鼠认知行为的影响 被引量:9

摘要 目的探讨茯苓对去卵巢大鼠认知行为的影响及其机制。方法大鼠双侧卵巢切除手术后随机分为茯苓低剂量治疗组(0.3g/kg)、中剂量治疗组(0.6g/kg)、高剂量治疗组(1.2g/kg),戊酸雌二醇组(0.8mg/kg)和模型对照组,每组10只,另设10只假手术组。模型组和假手术组灌胃0.9%氯化钠溶液。灌胃给药5周后Morris水迷宫法检测学习记忆能力的变化,Western blotting法检测BDNF和TrkB蛋白及其磷酸化表达水平。结果中、高剂量茯苓组和戊酸雌二醇组可以显著缩短大鼠逃避潜伏期,提高穿越站台次数,上调BDNF蛋白及TrkB蛋白磷酸化的表达水平,差异具有统计学意义(P<0.05)。结论茯苓能够通过激活BDNF/TrkB信号通路,促进去卵巢大鼠学习、记忆能力,改善认知行为。 Objective To observe the effect of Poria cocos on cognitional function in ovariectomized rats and study the possible mechanism.Methods Ovariectomized rats were randomly divided into five groups:high-,medium-,low-dose Poria cocos group,estradiol valerate group,model control group and sham operation group(n=10).The rats in the model control group and sham operation group were given 10 ml/kg 0.9%sodium chloride solution.The rats in the Estradiol valerate group were given 0.8 mg/kg estradiol valerate.The rats in the high-,medium-,low-dose Poria cocos group were given 1.2,0.6,0.3g/kg Poria cocos,respectively.Five weeks after the treatment,orris water maze test was used to detect learning and memory ability of rats,and western-blot was used to detect expression abundance of BDNF and TrkB protein in hippocampal tissue of rats.Results The high-,medium-Poria cocos group and estradiol valerate group could significantly reduce the escape latency of ovariectomized rats,increase the number of crossing plat-forms and effective residence time,and up-regulate the phosphorylation of BDNF and TrkB protein,the difference was statistically significant when compared with the model group(P<0.05).Conclusion Poria cocos can promote the learning and memory ability of ovariectomized rats by activating BDNF/TrkB signaling pathway.
